@article{fdi:010096297, title = {{R}ediscovering the lost : e{DNA} detection of freshwater decapods on {R}{\'e}union {I}sland}, author = {{J}annel, {L}. {A}. and de {M}azancourt, {V}. and {K}eith, {P}. and {J}ourand, {P}hilippe and {V}alade, {P}.}, editor = {}, language = {{ENG}}, abstract = {{F}reshwater ecosystems on tropical oceanic islands host unique biodiversity. {O}n {R}{\'e}union {I}sland, freshwater decapod crustaceans are key indicators of ecosystem health, yet their distribution and conservation status remain poorly documented. {T}his study aimed to investigate the presence of the endemic prawn {M}acrobrachium hirtimanus, historically reported but unconfirmed for decades, while producing the first island-wide molecular inventory of freshwater decapods using environmental {DNA} (e{DNA}) metabarcoding. {W}ater samples (4 {L}) were collected from 19 riverine sites, and e{DNA} was extracted and amplified using mitochondrial 16{S} r{RNA} primers validated for crustacean detection, followed by high-throughput sequencing. {B}ioinformatics analyses yielded 250,545 reads and 198 operational taxonomic units ({OTU}s), representing diverse amphidromous freshwater, coastal marine, and terrestrial decapods. {S}pecies richness was largely uniform across sites, though {R}ivi{\`e}re des {R}oches, {S}ainte-{M}arie, and {S}aint-{D}enis hosted higher diversity. {B}eta diversity showed strong spatial structuring among watersheds, driven primarily by species turnover. {W}idespread taxa included {A}tyoida serrata, {C}aridina typus, and {M}acrobrachium australe, while {C}. henriettae and {H}alocaridinides sp. were more localized. {M}. hirtimanus was not detected, consistent with its current absence, though continued targeted surveys are required before any formal reassessment. {O}verall, this study demonstrates the utility of e{DNA} metabarcoding for monitoring freshwater biodiversity and informing conservation planning on tropical islands.}, keywords = {{D}ecapod ; environmental {DNA} ; freshwater ; tropical island ; amphidromous ; {REUNION}}, booktitle = {}, journal = {{K}nowledge and {M}anagement of {A}quatic {E}cosystems}, numero = {427}, pages = {2 [13 p.]}, ISSN = {1961-9502}, year = {2026}, DOI = {10.1051/kmae/2025035}, URL = {https://www.documentation.ird.fr/hor/fdi:010096297}, }
