@article{fdi:010093545, title = {{I}dentification of a susceptibility gene for {X}anthomonas oryzae pv. oryzae in {V}ietnamese elite rice ({O}ryza sativa {L}.) cultivar {B}acthom 7}, author = {{P}huong, {N}. {D}. and {G}ioi, {D}. {H}. and {Q}uyen, {C}. {L}. and {H}ung, {N}. {X}. and {Y}en, {H}. {H}. and {H}uong, {B}. {T}. {T}. and {C}unnac, {S}{\'e}bastien and {X}uan, {T}. {D}. and {H}oi, {P}. {X}.}, editor = {}, language = {{ENG}}, abstract = {{X}anthomonas oryzae pv. oryzae ({X}oo) is the causal agent of bacterial leaf blight ({BLB}) disease in rice, exerting a detrimental impact on the yield of various rice cultivars in {V}ietnam, notably the {B}acthom 7 ({BT}7) variety. {X}oo possesses transcription activator-like effectors ({TALE}s) which modulate the expression of host genes by specifically binding to the effector-binding element ({EBE}) sequence in target promoter regions, thereby facilitating bacterial proliferation and pathogenesis on rice plants. {N}otably, {O}s{SWEET}13, a member of the {S}ugar {W}ill {E}ventually be {E}xported {T}ransporter ({SWEET}) gene family encoding sugar transporter proteins, has been identified as a prominent susceptibility ({S}) gene for {BLB}. {T}his investigation focuses on elucidating the role of {O}s{SWEET}13 in the susceptibility of {BT}7 to {BLB} disease in {V}ietnam. {V}ietnamese {X}oo ({VXO}) strains were observed to elicit characteristic {BLB} symptoms on {BT}7 and trigger the expression of {O}s{SWEET}13 during the course of infection. {T}o further probe this, a clustered regularly interspaced palindromic repeats/{CRISPR}-associated protein 9 ({CRISPR}/{C}as9) construct targeting the {EBE} {P}th{X}o2{A} site on the {O}s{SWEET}13 promoter was designed and introduced into {BT}7 rice plants through {A}grobacterium tumefaciens, resulting in the creation of rice lines with {O}s{SWEET}13 promoter modifications. {U}pon conducting detailed genotype and phenotype analyses of the gene-edited rice plants, it was ascertained that the mutation exhibited no adverse effects on the principal agronomic traits under scrutiny. {N}otably, a transgene-free homozygous 3-nucleotide deletion modified rice line displayed diminished {O}s{SWEET}13 gene expression and a significant reduction in {BLB} development upon inoculation with {VXO} strains. {T}his study unequivocally establishes {O}s{SWEET}13 as an {S} gene for {VXO}, underscoring its significance in breeding programs aimed at enhancing {BLB} resistance in the {BT}7 variety through the utilization of {CRISPR}/{C}as9 technology.}, keywords = {{B}ac thom 7 ; {CRISPR}/{C}as9 ; {O}s{SWEET}13 ; {P}th{X}o2{A} ; {X}anthomonas ; {VIET} {NAM}}, booktitle = {}, journal = {{P}lant {B}reeding}, volume = {[{E}arly access]}, numero = {}, pages = {[10 p.]}, ISSN = {0179-9541}, year = {2025}, DOI = {10.1111/pbr.13283}, URL = {https://www.documentation.ird.fr/hor/fdi:010093545}, }
