@article{fdi:010092006, title = {{U}ncovering the physiology and distribution of thallium in {T}l-hyperaccumulating and {T}l-sensitive populations of {B}iscutella laevigata {L}}, author = {{S}alinitro, {M}. and {I}snard, {S}andrine and {B}rueckner, {D}. and {S}piers, {K}. {M}. and {A}arts, {M}. {G}. {M}. and {R}emigio, {A}. {C}. and van der {E}nt, {A}.}, editor = {}, language = {{ENG}}, abstract = {{T}hallium ({T}l) is extremely toxic to all life forms and is an emerging pollutant. {P}lants in the {B}rassicaceae family, including edible crops, have an enhanced capacity for {T}l accumulation, even from soils with low thallium concentration. {T}he most extreme {T}l hyperaccumulator is {B}iscutella laevigata, capable of attaining >32 000 mu g {T}l g(-1) dry weight ({DW}) in its leaves. center dot {M}ethods {B}iscutella laevigata from a non-metallicolous accession ({F}eltre, {I}taly) and a metallicolous accession ({L}es {M}alines, {F}rance) were subjected to a dosing experiment in hydroponics (0, 5 and 30 mu m {T}l), followed by synchrotron-based micro-{X}-ray fluorescence analysis to elucidate tissue- and cellular-level {T}l distribution. center dot {K}ey {R}esults {F}low cytometric data on the two accessions showed that the {F}eltre accession has a genome size twice of that of the {L}es {M}alines accession (256 and 125 pg per 2{C}, respectively), suggesting that they are phylogenetically distant populations. {T}he {F}eltre accession did not accumulate {T}l (125 mu g {T}l g(-1) {DW} on average in leaves) at the 5 mu m {T}l dose level, whereas the {L}es {M}alines accession had a mean of 1750 mu g {T}l g(-1) {DW}, with peaks of 24 130 mu g {T}l g(-1) {DW}, at the 30 mu m {T}l dose level. {A}t 30 mu m {T}l, the non-metallicolous accession did not grow, and at 5 mu m {T}l it showed reduced biomass compared with the metallicolous one. {I}n the {L}es {M}alines accession, the synchrotron-based micro-{X}-ray fluorescence analysis revealed that {T}l was localized in the vacuoles of epidermal cells, especially underneath trichomes and in trichome basal cells. {T}hallium also occurred in solid crystalline deposits (3-5 mu m in size, similar to 40 wt% {T}l) that were found mainly in foliar margins and under trichome bases. center dot {C}onclusions {B}iscutella laevigata is an attractive model for studying {T}l hypertolerance and hyperaccumulation on account of the extreme expression of this trait and its marked intraspecific variability.}, keywords = {{B}ioconcentration ; phytoextraction ; intraspecific variation ; metal tolerance ; synchrotron-based micro-{X}-ray fluorescence ; {B}iscutella laevigata {L} ; {FRANCE}}, booktitle = {}, journal = {{A}nnals of {B}otany}, volume = {134}, numero = {}, pages = {803--814}, ISSN = {0305-7364}, year = {2024}, DOI = {10.1093/aob/mcae115}, URL = {https://www.documentation.ird.fr/hor/fdi:010092006}, }
