@article{fdi:010090056, title = {45{S} r{DNA} diversity in natura as one step towards ribosomal heterogeneity in {A}rabidopsis thaliana}, author = {{D}elorme-{H}inoux, {V}. and {M}bodj, {A}ssane and {B}rando, {S}. and {D}e {B}ures, {A}. and {L}lauro, {C}. and {C}ovato, {F}. and {G}arrigue, {J}. and {G}uisset, {C}. and {B}orrut, {J}. and {M}irouze, {M}arie and {R}eichheld, {J}. {P}. and {S}aez-{V}aasquez, {J}.}, editor = {}, language = {{ENG}}, abstract = {{T}he keystone of ribosome biogenesis is the transcription of 45{S} r{DNA}. {T}he {A}rabidopsis thaliana genome contains hundreds of 45{S} r{DNA} units; however, they are not all transcribed. {N}otably, 45{S} r{DNA} units contain insertions/deletions revealing the existence of heterogeneous r{RNA} genes and, likely, heterogeneous ribosomes for r{RNA}s. {I}n order to obtain an overall picture of 45{S} r{DNA} diversity sustaining the synthesis of r{RNA}s and, subsequently, of ribosomes in natura, we took advantage of 320 new occurrences of {A}rabidopsis thaliana as a metapopulation named {A}t66, sampled from 0 to 1900 m of altitude in the eastern {P}yrenees in {F}rance. {W}e found that the 45{S} r{DNA} copy number is very dynamic in natura and identified new genotypes for both 5' and 3' {E}xternal {T}ranscribed {S}pacers ({ETS}). {I}nterestingly, the highest 5'{ETS} genotype diversity is found in altitude while the highest 3'{ETS} genotype diversity is found at sea level. {S}tructural analysis of 45{S} r{DNA} also shows conservation in natura of specific 5'{ETS} and 3'{ETS} sequences/features required to control r{DNA} expression and the processing of r{RNA}s. {I}n conclusion, {A}t66 is a worthwhile natural laboratory, and unraveled 45{S} r{DNA} diversity represents an interesting starting material to select subsets for r{DNA} transcription and alter the r{RNA} composition of ribosomes both intra- and inter-site.}, keywords = {r{DNA} ; {A}rabidopsis thaliana ; brassicaceae ; ribosomal heterogeneity ; natural variation}, booktitle = {}, journal = {{P}lants}, volume = {12}, numero = {14}, pages = {2722 [21 p.]}, year = {2023}, DOI = {10.3390/plants12142722}, URL = {https://www.documentation.ird.fr/hor/fdi:010090056}, }
