@article{fdi:010090010, title = {{G}enetic factors for differentiated thyroid cancer in {F}rench {P}olynesia : new candidate loci}, author = {{Z}idane, {M}. and {H}aber, {M}. and {T}ruong, {T}. and {R}achedi, {F}. and {O}ry, {C}. and {C}hevillard, {S}. and {B}lanche, {H}. and {O}laso, {R}. and {B}oland, {A}. and {C}onte, {E}. and {K}arimi, {M}. and {R}en, {Y}. and {X}haard, {C}. and {S}ouchard, {V}. and {G}ardon, {J}acques and {T}aquet, {M}arc and {B}ouville, {A}. and {D}eleuze, {J}. {F}. and {D}rozdovitch, {V}. and de {V}athaire, {F}. and {C}azier, {J}. {B}.}, editor = {}, language = {{ENG}}, abstract = {{B}ackground {P}opulations of {F}rench {P}olynesia ({FP}), where {F}rance performed atmospheric tests between 1966 and 1974, experience a high incidence of differentiated thyroid cancer ({DTC}). {H}owever, up to now, no sufficiently large study of {DTC} genetic factors in this population has been performed to reach definitive conclusion. {T}his research aimed to analyze the genetic factors of {DTC} risk among the native {FP} populations. {M}ethods {W}e analyzed more than 300 000 single nucleotide polymorphisms ({SNP}s) genotyped in 283 {DTC} cases and 418 matched controls born in {FP}, most being younger than 15 years old at the time of the first nuclear tests. {W}e analyzed the genetic profile of our cohort to identify population subgroups. {W}e then completed a genome-wide analysis study on the whole population. {R}esults {W}e identified a specific genetic structure in the {FP} population reflecting admixture from {A}sian and {E}uropean populations. {W}e identified three regions associated with increased {DTC} risk at 6q24.3, 10p12.2, and 17q21.32. {T}he lead {SNP}s at these loci showed respective p-values of 1.66 x 10(-7), 2.39 x 10(-7,) and 7.19 x 10(-7) and corresponding odds ratios of 2.02, 1.89, and 2.37. {C}onclusion {O}ur study results suggest a role of the loci 6q24.3, 10p12.2 and 17q21.32 in {DTC} risk. {H}owever, a whole genome sequencing approach would be better suited to characterize these factors than genotyping with microarray chip designed for the {C}aucasian population. {M}oreover, the functional impact of these three new loci needs to be further explored and validated.}, keywords = {differentiated thyroid cancer ; population genetics ; genetic ; susceptibility ; {POLYNESIE} {FRANCAISE}}, booktitle = {}, journal = {{P}recision {C}linical {M}edicine}, volume = {6}, numero = {2}, pages = {ad015 [10 p.]}, ISSN = {2096-5303}, year = {2023}, DOI = {10.1093/pcmedi/pbad015}, URL = {https://www.documentation.ird.fr/hor/fdi:010090010}, }
