@article{fdi:010086188, title = {{B}iological and molecular characterization of the {C}ucurbit aphid-borne yellows virus affecting {C}ucurbits in {T}unisia}, author = {{M}nari-{H}attab, {M}. and {G}authier, {N}athalie and {Z}ouba, {A}.}, editor = {}, language = {{ENG}}, abstract = {{S}urveys of yellowing viruses under nonheated and geothermal heated plastic tunnels and in open field crops of melon ({C}ucumis melo), cucumber ({C}. sativus), zucchini ({C}ucurbita pepo), squash ({C}. maxima), watermelon ({C}itrullus lanatus), and ware cucurbit ({E}cballium elaterium) were carried out year-round during 2000-2001, 2003, and 2004 in the major cucurbit-growing areas in {T}unisia. {S}evere yellowing symptoms on older leaves of cucurbits were observed in open fields and under plastic-tunnel production systems. {T}hese yellowing symptoms and large populations of aphids ({A}phis gossypii) on a diversity of cucurbit crops in {T}unisia support the hypothesis of a viral cause of the disease. {V}irus identification using double-antibody sandwich enzyme-linked immunosorbent assay ({DAS}-{ELISA}), followed by reverse transcription-polymerase chain reaction ({RT}-{PCR}) and immunocapture ({IC})-{RT}-{PCR} showed that {C}ucurbit aphid-borne yellows virus ({CABYV}) was largely distributed in melon, cucumber, zucchini, squash, and watermelon crops. {W}are cucurbit ({E}. elaterium) and lettuce ({L}actuca sativa) crops were identified as potential {CABYV} reservoirs. {T}he {RT}-{PCR}-amplified partial coat protein ({CP}) and {P}4 genes were cloned and sequenced from nine {T}unisian {CABYV} isolates. {CP} and {P}4 gene nucleotide and amino acid sequence comparisons as well as phylogenetic reconstructions showed that the {T}unisian isolates clustered into two major subgroups. {C}omparisons with {CABYV} sequences retrieved from {G}en{B}ank showed high nucleotide and {CP} amino acid identities, and close relationships of the {T}unisian isolates with {I}talian and {F}rench isolates.}, keywords = {}, booktitle = {}, journal = {{P}lant {D}isease}, volume = {93}, numero = {10}, pages = {1065--1072}, ISSN = {0191-2917}, year = {2009}, DOI = {10.1094/pdis-93-10-1065}, URL = {https://www.documentation.ird.fr/hor/fdi:010086188}, }
