@article{fdi:010086037, title = {{M}olecular characterization of four {M}exican isolates of {T}rypanosoma cruzi and their profile susceptibility to nifurtimox}, author = {{J}aime, {L}. {D}. and {A}racely, {L}. {M}. and {P}aulina, {O}. {M}. and {D}umonteil, {E}. and {B}arnab{\'e}, {C}hristian and {W}aleckx, {E}tienne and {H}ernandez-{G}iles, {R}. {G}. and {R}amos-{L}igonio, {A}.}, editor = {}, language = {{ENG}}, abstract = {{P}urpose {T}he objective of this study was to molecularly characterize {M}exican isolates of {T}. cruzi obtained from infected triatomine bugs (the vectors of {T}. cruzi) and to evaluate their susceptibility to {N}ifurtimox ({NFX}). {M}ethods {T}hree isolates obtained from {T}riatoma dimidiata (collected in the {S}tate of {V}eracruz) and one isolate obtained from {T}riatoma bassolsae (collected in the {S}tate of {P}uebla) were molecularly characterized and the expression of genes associated with natural resistance to {NFX} was analyzed by q{PCR}. {R}esults {M}olecular characterization by {PCR} showed that isolates {Z}n3, {Z}n5, and {SRB}1 belong to the {DTU} {T}c{I}, while isolate {S}um3 belongs to {T}c{IV}. {T}he latter was also confirmed by sequencing of mitochondrial genes. {I}solate {Z}n5 was the most sensitive to treatment with {NFX} ({IC}50, 6.8 mu {M}), isolates {SRB}1 and {Z}n3 were partially resistant ({IC}50, 12.8 mu {M} and 12.7 mu {M}) and isolate {S}um3 showed a high degree of resistance to {NFX} ({IC}50, 21.4 mu {M}). {W}e also found an association between decreased {NTR}1 or {OYE} gene expression with {NFX} resistance. {C}onclusion {O}ur results also evidenced a high variability in the susceptibility to {NFX} of these {T}. cruzi isolates {C}entral and {S}outheastern {M}exico, suggesting the presence of naturally resistant isolates circulating in the country. {T}hese results have important implications for defining treatment policies for patients with {C}hagas disease.}, keywords = {{C}hagas disease ; {T}rypanosoma cruzi ; {M}exican isolates ; {DTU}s ; {N}ifurtimox ; {R}esistance genes ; {V}eracruz ; {P}uebla ; {MEXIQUE} ; {VERACRUZ} ; {PUEBLA}}, booktitle = {}, journal = {{A}cta {P}arasitologica}, volume = {[{E}arly access]}, numero = {}, pages = {[10 p.]}, ISSN = {1230-2821}, year = {2022}, DOI = {10.1007/s11686-022-00608-3}, URL = {https://www.documentation.ird.fr/hor/fdi:010086037}, }
