@article{fdi:010083878, title = {{I}mproved bacterial leaf blight disease resistance in the major elite {V}ietnamese rice cultivar {TBR}225 via editing of the {O}s{SWEET}14 promoter}, author = {{D}uy, {P}. {N}. and {L}an, {D}. {T}. and {T}hu, {H}. {P}. and {T}hu, {H}. {P}. {T}. and {T}hanh, {H}. {N}. and {P}ham, {N}. {P}. and {A}uguy, {F}lorence and {T}hu, {H}. {B}. {T}. and {M}anh, {T}. {B}. and {C}unnac, {S}{\'e}bastien and {P}ham, {X}. {H}.}, editor = {}, language = {{ENG}}, abstract = {{TBR}225 is one of the most popular commercial rice varieties in {N}orthern {V}ietnam. {H}owever, this variety is highly susceptible to bacterial leaf blight ({BLB}), a disease caused by {X}anthomonas oryzae pv. oryzae ({X}oo) which can lead to important yield losses. {O}s{SWEET}14 belongs to the {SWEET} gene family that encodes sugar transporters. {T}ogether with other {C}lade {III} members, it behaves as a susceptibility ({S}) gene whose induction by {A}sian {X}oo {T}ranscription-{A}ctivator-{L}ike {E}ffectors ({TALE}s) is absolutely necessary for disease. {I}n this study, we sought to introduce {BLB} resistance in the {TBR}225 elite variety. {F}irst, two {V}ietnamese {X}oo strains were shown to up-regulate {O}s{SWEET}14 upon {TBR}225 infection. {T}o investigate if this induction is connected with disease susceptibility, nine {TBR}225 mutant lines with mutations in the {A}vr{X}a7, {P}th{X}o3 or {T}al{F} {TALE}s {DNA} target sequences of the {O}s{SWEET}14 promoter were obtained using the {CRISPR}/{C}as9 editing system. {G}enotyping analysis of {T}-0 and {T}-1 individuals showed that mutations were stably inherited. {N}one of the examined agronomic traits of three transgene-free {T}2 edited lines were significantly different from those of wild-type {TBR}225. {I}mportantly, one of these {T}-2 lines, harboring the largest homozygous 6-bp deletion, displayed decreased {O}s{SWEET}14 expression as well as a significantly reduced susceptibility to a {V}ietnamese {X}oo strains and complete resistance to another one. {O}ur findings indicate that {CRISPR}/{C}as9 editing conferred an improved {BLB} resistance to a {V}ietnamese commercial elite rice variety.}, keywords = {{VIET} {NAM}}, booktitle = {}, journal = {{PL}o{S} {O}ne}, volume = {16}, numero = {9}, pages = {e0255470 [16 p.]}, ISSN = {1932-6203}, year = {2021}, DOI = {10.1371/journal.pone.0255470}, URL = {https://www.documentation.ird.fr/hor/fdi:010083878}, }
