@article{fdi:010083650, title = {{I}n-house {HIV}-1 {RNA} real-time {RT}-{PCR} assays : principle, available tests and usefulness in developing countries}, author = {{R}ouet, {F}. and {M}{\'e}nan, {H}. and {V}iljoen, {J}. and {N}go-{G}iang-{H}uong, {N}icole and {M}andaliya, {K}. and {V}al{\'e}a, {D}. and {L}ien, {T}. {X}. and {D}anaviah, {S}. and {R}ousset, {D}. and {G}anon, {A}. and {N}errienet, {E}.}, editor = {}, language = {{ENG}}, abstract = {{T}he principle of currently available licensed {HIV}-1 {RNA} assays is based on real-time technologies that continuously monitor the fluorescence emitted by the amplification products. {B}esides these assays, in-house quantitative (q) real-time reverse transcription ({RT})-{PCR} ({RT}-q{PCR}) tests have been developed and evaluated particularly in developing countries, for two main reasons. {F}irst, affordable and generalized access to {HIV}-1 {RNA} viral load is urgently needed in the context of expected universal access to prevention and antiretroviral treatment programs in these settings. {S}econd, since many non-{B} subtypes, circulating recombinant forms and unique recombinant forms circulate in these areas, in-house {HIV}-1 {RNA} {RT}-q{PCR} assays are ideal academic tools to thoroughly evaluate the impact of {HIV}-1 genetic diversity on the accuracy of {HIV}-1 {RNA} quantification, as compared with licensed techniques. {T}o date, at least 15 distinct in-house assays have been designed. {T}hey differ by their chemistry and the {HIV}-1 target sequence (located in gag, {P}ol-{IN} or {LTR} gene). {A}nalytical performances of the tests that have been extensively evaluated appear at least as good as (or even better than) those of approved assays, with regard to {HIV}-1 strain diversity. {T}heir clinical usefulness has been clearly demonstrated for early diagnosis of pediatric {HIV}-1 infection and monitoring of highly active antiretroviral therapy efficacy. {T}he {LTR}-based {HIV}-1 {RNA} {RT}-q{PCR} assay has been evaluated by several groups under the auspices of the {A}gence {N}ationale de {R}echerches sur le {SIDA} et les hepatites virales {B} et {C}. {I}t exists now as a complete standardized commercial test.}, keywords = {{D}eveloping country ; {G}enetic diversity ; {H}iv-1 ; {P}cr ; {Q}uantitative ; {R}eal-time ; {R}everse transcription ; {R}na ; {V}iral load}, booktitle = {}, journal = {{E}xpert {R}eview of {M}olecular {D}iagnostics}, volume = {8}, numero = {5}, pages = {635--650}, ISSN = {1473-7159}, year = {2008}, DOI = {10.1586/14737159.8.5.635}, URL = {https://www.documentation.ird.fr/hor/fdi:010083650}, }
