@article{fdi:010077595, title = {{D}evelopment of a variable number of tandem repeats typing scheme for the bacterial rice pathogen {X}anthomonas oryzae pv. oryzicola}, author = {{Z}hao, {S}. and {P}oulin, {L}ucie and {R}odriguez-{R}, {L}.{M}. and {S}erna, {N}.{F}. and {L}iu, {S}. {Y}. and {W}onni, {I}. and {S}zurek, {B}oris and {V}erdier, {V}al{\'e}rie and {L}each, {J}. {E}. and {H}e, {Y}. {Q}. and {F}eng, {J}. {X}. and {K}oebnik, {R}alf}, editor = {}, language = {{ENG}}, abstract = {{X}anthomonas oryzae pv. oryzicola is an important bacterial pathogen responsible for outbreaks of bacterial leaf streak ({BLS}) on rice, mostly occurring in {A}sia and parts of {A}frica. {T}o better monitor epidemics and assess population structures, efficient tools that allow the precise identification and diagnosis of pathogenic populations are needed. {I}n this study, we explored variable numbers of tandem repeats ({VNTR}) as a fast, reliable, and cost-effective molecular typing tool. {S}creening of three {X}. oryzae pv. oryzicola genome sequences ({P}hilippine strain {BLS}256, {C}hinese strain {GX}01, and {M}alian strain {MAI}10) predicted 28 candidate {VNTR} loci. {P}rimer pairs for polymerase chain reaction ({PCR}) amplification of all 28 loci were designed and applied to a panel of 20 {X}. oryzae pv. oryzicola strains originating from {A}sia and {A}frica. {S}equencing of {PCR} amplicons revealed 25 robust and polymorphic {VNTR} loci that are shared among {A}sian and {A}frican {X}. oryzae pv. oryzicola strains. {A} dendrogram constructed from 25 {VNTR} loci indicated that most {A}sian strains are clearly discriminated from {A}frican strains. {H}owever, in agreement with previous reports, one strain from {M}ali is related to {A}sian strains, pointing to a possible introduction of {A}sian strains to the {A}frican continent. {T}he new {VNTR}-based tool described here is useful for studies of population structures and epidemiological monitoring of {X}. oryzae pv. oryzicola.}, keywords = {{MLVA} ; {AFRIQUE} ; {ASIE}}, booktitle = {}, journal = {{P}hytopathology}, volume = {102}, numero = {10}, pages = {948--956}, ISSN = {0031-949{X}}, year = {2012}, DOI = {10.1094/phyto-04-12-0078-r}, URL = {https://www.documentation.ird.fr/hor/fdi:010077595}, }
