@article{fdi:010073788, title = {{P}rotists within corals : the hidden diversity}, author = {{C}lerissi, {C}. and {B}runet, {S}. and {V}idal-{D}upiol, {J}. and {A}djeroud, {M}ehdi and {L}epage, {P}. and {G}uillou, {L}. and {E}scoubas, {J}. {M}. and {T}oulza, {E}.}, editor = {}, language = {{ENG}}, abstract = {{P}revious observations suggested that microbial communities contribute to coral health and the ecological resilience of coral reefs. {H}owever, most studies of coral microbiology focused on prokaryotes and the endosymbiotic algae {S}ymbiodinium. {I}n contrast, knowledge concerning diversity of other protists is still lacking, possibly due to methodological constraints. {A}s most eukaryotic {DNA} in coral samples was derived from hosts, protist diversity was missed in metagenome analyses. {T}o tackle this issue, we designed blocking primers for {S}cleractinia sequences amplified with two primer sets that targeted variable loops of the 18{S} r{RNA} gene (18{SV}1{V}2 and 18{SV}4). {T}hese blocking primers were used on environmental colonies of {P}ocillopora damicornis sensu lato from two regions with contrasting thermal regimes ({D}jibouti and {N}ew {C}aledonia). {I}n addition to {S}ymbiodinium clades {A}/{C}/{D}, {L}icnophora and unidentified coccidia genera were found in many samples. {I}n particular, coccidian sequences formed a robust monophyletic clade with other protists identified in {A}garicia, {F}avia, {M}ontastraea, {M}ycetophyllia, {P}orites, and {S}iderastrea coral colonies. {M}oreover, {L}icnophora and coccidians had different distributions between the two geographic regions. {A} similar pattern was observed between {S}ymbiodinium clades {C} and {A}/{D}. {A}lthough we were unable to identify factors responsible for this pattern, nor were we able to confirm that these taxa were closely associated with corals, we believe that these primer sets and the associated blocking primers offer new possibilities to describe the hidden diversity of protists within different coral species.}, keywords = {holobiont ; protists ; symbiosis ; metabarcoding ; blocking primer ; {S}cleractinia ; {P}ocillopora damicornis ; {DJIBOUTI} ; {NOUVELLE} {CALEDONIE}}, booktitle = {}, journal = {{F}rontiers in {M}icrobiology}, volume = {9}, numero = {}, pages = {art. 2043 [13 p.]}, ISSN = {1664-302{X}}, year = {2018}, DOI = {10.3389/fmicb.2018.02043}, URL = {https://www.documentation.ird.fr/hor/fdi:010073788}, }
