@article{fdi:010072755, title = {{P}athogenicity and phylogenetic analysis of {C}lavibacter michiganensis strains associated with tomato plants in {I}ran}, author = {{O}sdaghi, {E}. and {A}nsari, {M}. and {T}aghavi, {S}. {M}. and {Z}arei, {S}. and {K}oebnik, {R}alf and {L}amichhane, {J}. {R}.}, editor = {}, language = {{ENG}}, abstract = {{D}uring 2013-2016, 277 tomato fields were surveyed across {I}ran to monitor the status of bacterial canker of tomato, caused by {C}lavibacter michiganensis subsp. michiganensis. {A}ltogether, 450 plant samples were collected, both with and without symptoms, from which 35 bacterial strains were recovered. {T}hese were positive for the {PCR} test performed using the {C}lavibacter-specific primer pair {CMR}16{F}1/{CMR}16{R}1. {B}ased on the phylogeny of the gyr{B} gene sequences, 31, three and one of the 35 strains were identified as {C}. michiganensis, {M}icrobacterium sp. and {A}grococcus sp., respectively. {T}he 31 strains of {C}. michiganensis were further identified as {C}. michiganensis subsp. michiganensis (23 strains), {C}. michiganensis subsp. tessellarius (six strains) and {C}lavibacter spp. (two strains). {T}his was subsequently confirmed by multilocus sequence analysis ({MLSA}) of five housekeeping genes (atp{D}, gyr{B}, ppk, rec{A} and rpo{B}). {I}n pathogenicity tests, all 23 strains induced wilting symptoms on tomato plants in greenhouse conditions, while no symptoms were observed on eggplant, bell pepper and chili pepper plants. {A}ll evaluated pathogenicity determinant genes (cel{A}, pat-1, tom{A}, ppa{A}, chp{C} and chp{G}) were detected in 18 out of 31 {C}. michiganensis strains, using eight specific primer pairs. {E}stimation of the number of nucleotide differences, sequence similarity matrix and {MLSA} clustered two peach-coloured strains ({T}om495 and {T}om532) separately from all nine previously described subspecies, thereby suggesting these two strains are a new subspecies of {C}. michiganensis. {H}owever, a detailed taxonomic study using multiphased molecular approaches is needed to delineate a formal taxonomic name for these atypical strains.}, keywords = {aetiology ; {MLST} ; solanaceous vegetables ; {IRAN}}, booktitle = {}, journal = {{P}lant {P}athology}, volume = {67}, numero = {4}, pages = {957--970}, ISSN = {0032-0862}, year = {2018}, DOI = {10.1111/ppa.12801}, URL = {https://www.documentation.ird.fr/hor/fdi:010072755}, }
