@article{fdi:010072438, title = {{C}ryptic genetic diversity in the mottled rabbitfish {S}iganus fuscescens with mitochondrial introgression at a contact zone in the {S}outh {C}hina {S}ea}, author = {{R}avago-{G}otanco, {R}. and de la {C}ruz, {T}. {L}. and {P}ante, {M}. {J}. and {B}orsa, {P}hilippe}, editor = {}, language = {{ENG}}, abstract = {{T}he taxonomy of the mottled rabbitfish {S}iganus fuscescens species complex has long been challenging. {I}n this study, we analyzed microsatellite genotypes, mitochondrial lineages, and morphometric data from 373 {S}. fuscescens individuals sampled from the northern {P}hilippines and {H}ong {K}ong ({S}outh {C}hina {S}ea, {P}hilippine {S}ea and {S}ulu {S}ea basins), to examine putative species boundaries in samples comprising three co-occurring mitochondrial lineages previously reported to characterize {S}. fuscescens ({C}lade {A} and {C}lade {B}) or {S}. canaliculatus ({C}lade {C}). {W}e report the existence of two cryptic species within {S}. fuscescens in the northeast region of the {S}outh {C}hina {S}ea and northern {P}hilippine {S}ea, supported by genetic and morphological differences. {I}ndividual-based assignment methods recovered concordant groupings of individuals into two nuclear genotype clusters ({C}luster 1, {C}luster 2) with (1) limited gene flow, if any, between them ({F}-{ST} = 0.241; {P} < 0.001); (2) low frequency of later-generation hybrids; (3) significant association with mitochondrial {C}lade {A} and {C}lade {B}, respectively; and (4) subtle yet significant body shape differences as inferred from geometric morphometric analysis. {T}he divergence between mitochondrial {C}lade {C} and the two other clades was not matched by genetic differences at microsatellite marker loci. {T}he occurrence of discordant mitonuclear combinations (20.5% of the total number of individuals) is thought to result from mitochondrial introgression, consistent with a scenario of demographic, and presumably spatial, post-{P}leistocene expansion of populations from northern regions into a secondary contact zone in the {S}outh {C}hina {S}ea. {M}itonuclear discordance due to introgression obscures phylogenetic relationships for recently-diverged lineages, and cautions against the use of mitochondrial markers alone for species identification within the mottled rabbitfish species complex in the {S}outh {C}hina {S}ea region.}, keywords = {{MER} {DE} {CHINE}}, booktitle = {}, journal = {{PL}o{S} {O}ne}, volume = {13}, numero = {2}, pages = {e0193220 [27 p.]}, ISSN = {1932-6203}, year = {2018}, DOI = {10.1371/journal.pone.0193220}, URL = {https://www.documentation.ird.fr/hor/fdi:010072438}, }
