@article{fdi:010061220, title = {{F}ive phylogenetically close rice {SWEET} genes confer {TAL} effector-mediated susceptibility to {X}anthomonas oryzae pv. oryzae}, author = {{S}treubel, {J}. and {P}esce, {C}. and {H}utin, {M}. and {K}oebnik, {R}alf and {B}och, {J}. and {S}zurek, {B}oris}, editor = {}, language = {{ENG}}, abstract = {{B}acterial plant-pathogenic {X}anthomonas strains translocate transcription activator-like ({TAL}) effectors into plant cells to function as specific transcription factors. {O}nly a few plant target genes of {TAL} effectors have been identified, so far. {T}hree plant {SWEET} genes encoding putative sugar transporters are known to be induced by {TAL} effectors from rice-pathogenic {X}anthomonas oryzae pv. oryzae ({X}oo). {W}e predict and validate that expression of {O}s{SWEET}14 is induced by a novel {TAL} effector, {T}al5, from an {A}frican {X}oo strain. {A}rtificial {TAL} effectors ({A}rt{TAL}s) were constructed to individually target 20 {SWEET} orthologs in rice. {T}hey were used as designer virulence factors to study which rice {SWEET} genes can support {X}oo virulence. {T}he {T}al5 target box differs from those of the already known {TAL} effectors {T}al{C}, {A}vr{X}a7 and {P}th{X}o3, which also induce expression of {O}s{SWEET}14, suggesting evolutionary convergence on key targets. {A}rt{TAL}s efficiently complemented an {X}oo tal{C} mutant, demonstrating that specific induction of {O}s{SWEET}14 is the key target of {T}al{C}. {A}rt{TAL}s that specifically target individual members of the rice {SWEET} family revealed three known and two novel {SWEET} genes to support bacterial virulence. {O}ur results demonstrate that five phylogenetically close {SWEET} proteins, which presumably act as sucrose transporters, can support {X}oo virulence.}, keywords = {designer {TALE} ; plant pathogen ; plant resistance ; rice ; {TALE} ; type-{III} ; effector ; virulence ; {O}s{SWEET}}, booktitle = {}, journal = {{N}ew {P}hytologist}, volume = {200}, numero = {3}, pages = {808--819}, ISSN = {1469-8137}, year = {2013}, DOI = {10.1111/nph.12411}, URL = {https://www.documentation.ird.fr/hor/fdi:010061220}, }
