@article{fdi:010060753, title = {{I}dentification of non-{TAL} effectors in {X}anthomonas oryzae pv. oryzae {C}hinese strain 13751 and analysis of their role in the bacterial virulence}, author = {{Z}hao, {S}huai and {M}o, {W}. {L}. and {W}u, {F}. and {T}ang, {W}. and {T}ang, {J}. {L}. and {S}zurek, {B}oris and {V}erdier, {V}al{\'e}rie and {K}oebnik, {R}alf and {F}eng, {J}. {X}.}, editor = {}, language = {{ENG}}, abstract = {{X}anthomonas oryzae pv. oryzae ({X}oo) is the causal agent of rice bacterial leaf blight, one of the most important rice bacterial diseases in {C}hina and many other countries. {T}he upstream portions of 41 candidate genes encoding non-transcription activator-like effectors of {X}oo {C}hinese strain 13751 were fused with the coding sequence of {A}vr{B}s1(59-445) in a broad host-range vector. {T}he constructed plasmids were respectively introduced into {X}oo strain 13751 and avr{B}s1 deletion mutant of {X}. campestris pv. campestris strain 8004 by tri-parental conjugation. {T}he resultant transconjugants were respectively tested for hypersensitive response ({HR}) elicitation on pepper {ECW}-10{R}. {N}ine strains were able to elicit {HR} on pepper, indicating that the nine genes ({XOO}0037, {XOO}0103, {XOO}0110, {XOO}0315, {XOO}1488, {XOO}2875, {XOO}3150, {XOO}3222 and {XOO}4134) encoded effectors. {A}mong them, xop{AE} (13751) ({XOO}0110), expressed in {X}oo strain 13751 growing in rice leaves, was a new experimentally confirmed effector gene. {X}op{AE}(13751) contains 11 leucine rich repeats. {F}urthermore, mutants for the nine effector genes were created in {X}oo strain 13751 and subsequently tested for virulence in rice. {A}s a result, only the xop{R} (13751) ({XOO}4134) deletion mutant {GXM}xop{R} showed a significant reduction in virulence in hybrid rice cv. {T}eyou63 compared to the wild type. {H}owever, the growth of {GXM}xop{R} in host plant rice was not affected. {T}hese results indicated that xop{R} (13751) was required for full virulence of {X}oo strain 13751 by inducing rice disease tolerance.}, keywords = {{B}acterial blight of rice ; {T}ype {III} secretion system ; {H}ypersensitive response ; {CHINE}}, booktitle = {}, journal = {{W}orld {J}ournal of {M}icrobiology and {B}iotechnology}, volume = {29}, numero = {4}, pages = {733--744}, ISSN = {0959-3993}, year = {2013}, DOI = {10.1007/s11274-012-1229-5}, URL = {https://www.documentation.ird.fr/hor/fdi:010060753}, }
