@article{fdi:010057066, title = {{I}nfluence of growth regulators on callogenesis and somatic embryo development in date palm ({P}hoenix dactylifera {L}.) sahelian cultivars}, author = {{S}an{\'e}, {D}. and {A}berlenc {B}ertossi, {F}r{\'e}d{\'e}rique and {D}iatta, {L}. {I}. {D}. and {G}ueye, {B}. and {D}aher, {A}. and {S}agna, {M}. and {D}uval, {Y}ves and {B}orgel, {A}lain}, editor = {}, language = {{ENG}}, abstract = {{T}his study provides a physiological analysis of somatic embryogenesis in four elite cultivars of date palms: {A}hmar, {A}msekhsi, {T}ijib, and {A}maside, from the initial callogenesis to establishment and proliferation of embryogenic suspension cultures. {S}omatic embryos development and in vitro plants rooting were also studied. {F}or each step, auxins and cytokinins concentrations were optimised. {T}he primary callogenesis from leaf explants of seedlings appeared highly dependent on genotype. {A}hmar (80%) and {A}msekhsi (76%) appeared highly callogenic, whereas {T}ijib (10%) and {A}maside (2%) produced low amounts of calluses. 2,4-{D}ichlorophenoxyacetic acid appeared favorable to the induction of primary callogenesis and its effect was enhanced by the addition of benzyl adenine or adenine sulfate. {S}econdary friable calli obtained fromchopped granular calli were used to initiate embryogenic cell suspensions inmedia supplied with 2,4-dichlorophenoxyacetic acid. {S}uspension cultures showed a growth rate of fourfold after four subcultures in presence of 2,4-dichlorophenoxyacetic acid 2mg/{L}. {O}ur results showed that a seven-day transitory treatment with benzyl adenine 0,5mg/{L} was necessary to optimize embryos development. {N}aphthalene acetic acid induced the development of primary orthogravitropic roots during embryos germination. {T}he comparison with cytofluorometry of nuclear {DNA} amounts showed no significant difference in ploidy level between regenerated plants and seedlings.}, keywords = {}, booktitle = {}, journal = {{S}cientific {W}orld {J}ournal}, numero = {}, pages = {837395}, ISSN = {1537-744{X}}, year = {2012}, DOI = {10.1100/2012/837395}, URL = {https://www.documentation.ird.fr/hor/fdi:010057066}, }
