@article{fdi:010055667, title = {{H}istorical contingencies modulate the adaptability of {R}ice {Y}ellow {M}ottle {V}irus}, author = {{P}oulicard, {N}ils and {G}alzi, {A}gn{\`e}s and {T}raore, {O}. and {V}ignols, {F}. and {G}hesqui{\`e}re, {A}lain and {K}onate, {G}. and {H}{\'e}brard, {E}ug{\'e}nie and {F}argette, {D}enis}, editor = {}, language = {{ENG}}, abstract = {{T}he rymv1-2 and rymv1-3 alleles of the {RYMV}1 resistance to {R}ice yellow mottle virus ({RYMV}), coded by an e{IF}(iso) 4{G}1 gene, occur in a few cultivars of the {A}siatic ({O}ryza sativa) and {A}frican ({O}. glaberrima) rice species, respectively. {T}he most salient feature of the resistance breaking ({RB}) process is the converse genetic barrier to rymv1-2 and rymv1-3 resistance breakdown. {T}his specificity is modulated by the amino acid (glutamic acid vs. threonine) at codon 49 of the {V}iral {P}rotein genome-linked ({VP}g), a position which is adjacent to the virulence codons 48 and 52. {I}solates with a glutamic acid ({E}) do not overcome rymv1-3 whereas those with a threonine ({T}) rarely overcome rymv1-2. {W}e found that isolates with {T}49 had a strong selective advantage over isolates with {E}49 in {O}. glaberrima susceptible cultivars. {T}his explains the fixation of the mutation {T}49 during {RYMV} evolution and accounts for the diversifying selection estimated at codon 49. {B}etter adapted to {O}. glaberrima, isolates with {T}49 are also more prone than isolates with {E}49 to fix rymv1-3 {RB} mutations at codon 52 in resistant {O}. glaberrima cultivars. {H}owever, subsequent genetic constraints impaired the ability of isolates with {T}49 to fix rymv1-2 {RB} mutations at codons 48 and 52 in resistant {O}. sativa cultivars. {T}he origin and role of the amino acid at codon 49 of the {VP}g exemplifies the importance of historical contingencies in the ability of {RYMV} to overcome {RYMV}1 resistance.}, keywords = {{AFRIQUE} {SUBSAHARIENNE} ; {MADAGASCAR}}, booktitle = {}, journal = {{P}los {P}athogens}, volume = {8}, numero = {1}, pages = {e1002482}, ISSN = {1553-7366}, year = {2012}, DOI = {10.1371/journal.ppat.1002482}, URL = {https://www.documentation.ird.fr/hor/fdi:010055667}, }
