@article{fdi:010053921, title = {{I}n vitro activity of iron-binding compounds against {S}enegalese isolates of {P}lasmodium falciparum}, author = {{P}radines, {B}. and {T}all, {A}. and {R}amiandrasoa, {F}.{R}. and {S}piegel, {A}. and {S}okhna, {C}heikh and {F}usai, {T}. and {M}osnier, {J}. and {D}aries, {W}. and {T}rape, {J}ean-{F}ran{\c{c}}ois and {K}unesch, {G}. and {P}arzy, {D}. and {R}ogier, {C}.}, editor = {}, language = {{ENG}}, abstract = {{O}bjectives: {T}he in vitro activities of {FR}160, a synthetic catecholate siderophore, and two iron-binding agents, desferrioxamine and doxycycline, were evaluated against {P}lasmodium falciparum isolates. {C}orrelations between these compounds and standard antimalarial drugs (chloroquine, quinine, amodiaquine, pyronarldine, artemether, artesunate, atovaquone, cycloguanil and pyrimethamine) were assessed to determine any degree of cross-resistance. {M}ethods: {B}etween {O}ctober 1997 and {F}ebruary 1998, and {S}eptember and {N}ovember 1998,189 {A} falciparum isolates were obtained in {D}ielmo and {N}diop ({D}akar). {T}heir susceptibilities were assessed using an isotopic, microwell format, drug susceptibility test. {R}esults: {T}he 137 inhibitory concentrations ({IC}50) values of {FR}160 ranged from 0.1 to 10 mu {M} and the geometric mean {IC}50 was 1.48 mu {M} (95% {C}l = 1.29-1.68 mu {M}). {T}he geometric mean {IC}50 of doxycycline for 121 isolates was 18.9 mu {M} (95% {C}l = 16.8-21.3 mu {M}) and that of desferrioxamine for 73 isolates was 20.7 mu {M} (95% {C}l = 17.3-24.8 mu {M}). {FR}160 was significantly less active against the chloroquine-resistant isolates ({P} < 0.0001). {T}he mean {IC}(50)s of doxycycline were significantly higher for the chloroquine-susceptible isolates than for the resistant parasites ({P} = 0.0447). {T}here was a weak correlation between the responses to {FR}160, desferrioxamine or doxycycline and those to the other antimalarial compounds (r(2) < 0.22). {C}onclusions: {T}he activities of {FR}160 and desferrioxamine, determined for {A} falciparum clones, were confirmed against 137 isolates. {T}he coefficients of determination between the responses to {FR}160, doxycycline or desferrioxamine and those to all the antimalarial drugs tested are too weak to suggest cross-resistance. {FR}160 could be a rationale partner to use in combination with doxycycline.}, keywords = {malaria ; iron chelation ; chemotherapy ; antimalarial drugs ; {S}enegal ; {SENEGAL}}, booktitle = {}, journal = {{J}ournal of {A}ntimicrobial {C}hemotherapy}, volume = {57}, numero = {6}, pages = {1093--1099}, ISSN = {0305-7453}, year = {2006}, DOI = {10.1093/jac/dkl117}, URL = {https://www.documentation.ird.fr/hor/fdi:010053921}, }
