@article{fdi:010053818, title = {{A}nopheles gambiae mosquito isolated neurons : a new biological model for optimizing insecticide/repellent efficacy}, author = {{L}avialle-{D}efaix, {C}. and {A}paire-{M}archais, {V}. and {L}egros, {C}. and {P}ennetier, {C}{\'e}dric and {M}ohamed, {A}. and {L}icznar, {P}. and {C}orbel, {V}incent and {L}apied, {B}.}, editor = {}, language = {{ENG}}, abstract = {{T}o understand better the mode of action of insecticides and repellents used in vector-borne diseases control, we developed a new biological model based on mosquito neurons isolated from adults {A}nopheles gambiae heads. {T}his cellular model is well adapted to multidisciplinary approaches: electrophysiology, pharmacology, molecular biology and biochemical assays. {U}sing {RT}-{PCR}, we demonstrated that isolated neurons express the nicotinic acetylcholine receptor subunit alpha 1 ({A}g alpha 1 n{A}ch{R}), two acetylcholinesterases ({AC}h{E}-1 and {AC}h{E}-2) and three voltage-gated ion channels required for membrane excitability ({A}g{C}av1, {A}g{N}av1 and {A}g{K}v1). {I}n order to correlate the expression of the different transcripts, encoding functional {A}g{N}av channel, n{AC}h{R} receptor and {AC}h{E} enzymes detected by {RT}-{PCR}, with electrophysiological activity we used patch-clamp technique. {W}e revealed that {A}g{N}av and {AC}h{E} which are targeted by insecticide and/or repellent were sensitive to the pyrethroid permethrin and to the repellent {DEET}, respectively. {I}n addition, using colorimetric method, we also showed that {AC}h{E} was sensitive to the carbamate propoxur. {T}hese results indicated that this novel neuronal mosquito model will lead to molecular and functional characterization of insecticide/repellent targets and appears as a powerful tool to investigate the development of highly specific and effective strategies for disease vector control.}, keywords = {{M}alaria ; {M}osquito ; {I}solated neurons ; {I}nsecticides ; {DEET} ; {V}ector-borne diseases}, booktitle = {}, journal = {{J}ournal of {N}euroscience {M}ethods}, volume = {200}, numero = {1}, pages = {68--73}, ISSN = {0165-0270}, year = {2011}, DOI = {10.1016/j.jneumeth.2011.06.003}, URL = {https://www.documentation.ird.fr/hor/fdi:010053818}, }
