@article{fdi:010052899, title = {{D}irect interaction between the {R}ice yellow mottle virus ({RYMV}) {VP}g and the central domain of the {R}ice e{IF}(iso)4{G}1 factor correlates with rice susceptibility and {RYMV} {V}irulence}, author = {{H}{\'e}brard, {E}ug{\'e}nie and {P}oulicard, {N}ils and {G}{\'e}rard, {C}. and {T}raor{\'e}, {O}. and {W}u, {H}. {C}. and {A}lbar, {L}aurence and {F}argette, {D}enis and {B}essin, {Y}. and {V}ignols, {F}.}, editor = {}, language = {{ENG}}, abstract = {{T}he adaptation of {R}ice yellow mottle virus ({RYMV}) to recessive resistance mediated by the rymv1-2 allele has been reported as a model to study the emergence and evolution of virulent variants. {T}he resistance and virulence factors have been identified as eukaryotic translation initiation factor e{IF}(iso)4{G}1 and viral genome linked protein ({VP}g), respectively, but the molecular mechanisms involved in their interaction are still unknown. {I}n this study, we demonstrated a direct interaction between {RYMV} {VP}g and the central domain of rice e{IF}(iso)4{G}1 both in vitro, using recombinant proteins, and in vivo, using a yeast two-hybrid assay. {I}nsertion of the {E}309{K} mutation in e{IF}(iso)4{G}1, conferring resistance in planta, strongly diminished the interaction with avirulent {VP}g. {T}he efficiency of the major virulence mutations at restoring the interaction with the resistance protein was assessed. {O}ur results explain the prevalence of virulence mutations fixed during experimental evolution studies and are consistent with the respective viral {RNA} accumulation levels of avirulent and virulent isolates. {O}ur results also explain the origin of the residual multiplication of wild-type isolates in rymv1-2 resistant plants and the role of genetic context in the poor adaptability of the {S}2/{S}3 strain. {F}inally, the strategies of {RYMV} and members of family {P}otyviridue to overcome recessive resistance were compared.}, keywords = {}, booktitle = {}, journal = {{M}olecular {P}lant-{M}icrobe {I}nteractions}, volume = {23}, numero = {11}, pages = {1506--1513}, ISSN = {0894-0282}, year = {2010}, DOI = {10.1094/mpmi-03-10-0073}, URL = {https://www.documentation.ird.fr/hor/fdi:010052899}, }
