@article{fdi:010052886, title = {{S}erodiagnosis of bovine trypanosomosis based on {HSP}70/{B}i{P} inhibition {ELISA}}, author = {{B}ossard, {G}. and {B}oulang{\'e}, {A}. and {H}olzmuller, {P}. and {T}h{\'e}venon, {S}. and {P}atrel, {D}elphine and {A}uthi{\'e}, {E}dith}, editor = {}, language = {{ENG}}, abstract = {{A}nimal trypanosomosis is a serious constraint to livestock productivity in tropical and subtropical countries. {T}he pathogenic trypanosomes in bovidae are {T}rypanosoma congolense, {T}. vivax, {T}. brucei and {T}. evansi. {C}urrent serological tests to detect trypanosome infections are based on the use of whole trypanosome lysates; their potential is limited by antigen instability, lack of reproducibility and lack of test specificity due to the antibody's long persistence after treatment. {T}he development of new tests based on recombinant technology that could be standardized and applied on a large scale at low cost would be very helpful. {T}he major invariant antigen recognized by {T}. congolense infected cattle belongs to the heat shock protein ({HSP}) 70 family and is closely related to mammalian {I}mmunoglobulin {B}inding {P}rotein ({B}i{P}). {T}o improve the initial {ELISA} based on a recombinant fragment of {HSP}70/{B}i{P}, we developed an inhibition {ELISA} using an anti-{B}i{P} monoclonal antibody and a full-length fusion protein expressed in {E}. coli. {H}ere we report on the development of the test and provide an initial assessment of its performance using sets of sera from experimental infections and from naturally infected cattle maintained in tsetse infested areas of {A}frica. {T}he {HSP}70/{BIP}-based inhibition {ELISA} shows a good sensitivity in cattle experimentally infected with {T}. congolense, with an improved sensitivity in secondary infections. {O}ne major advantage, particularly for its further application in national laboratories, is that one single set of reagents and one single procedure are sufficient to apply on different mammalian host species infected with different trypanosome species.}, keywords = {{S}erodiagnosis ; {I}nhibition {ELISA} ; {B}ovine trypanosomosis ; {HSP}70/{B}i{P}}, booktitle = {}, journal = {{V}eterinary {P}arasitology}, volume = {173}, numero = {1-2}, pages = {39--47}, ISSN = {0304-4017}, year = {2010}, DOI = {10.1016/j.vetpar.2010.06.016}, URL = {https://www.documentation.ird.fr/hor/fdi:010052886}, }
