@article{fdi:010049531, title = {{L}arge-scale transposon mutagenesis of photosynthetic {B}radyrhizobium {S}p strain {ORS}278 reveals new genetic loci putatively important for nod-independent symbiosis with {A}eschynomene indica}, author = {{B}onaldi, {K}. and {G}ourion, {B}. and {F}ardoux, {J}o{\¨e}l and {H}annibal, {L}aure and {C}artieaux, {F}abienne and {B}oursot, {M}arc and {V}allenet, {D}. and {C}haintreuil, {C}l{\'e}mence and {P}rin, {Y}. and {N}ouwen, {N}ico and {G}iraud, {E}ric}, editor = {}, language = {{ENG}}, abstract = {{P}hotosynthetic {B}radyrhizobium strains possess the unusual ability to form nitrogen-fixing nodules on a specific group of legumes in the absence of {N}od factors. {T}o obtain insight into the bacterial genes involved in this {N}od-independent symbiosis, we screened 15,648 {T}n5 mutants of {B}radyrhizobium sp. strain {ORS}278 for clones affected in root symbiosis with {A}eschynomene indica. {F}rom the 268 isolated mutants, 120 mutants were altered in nodule development ({N}dv(-)) and 148 mutants were found to be deficient in nitrogen fixation ({F}ix(-)). {M}ore than 50% of the {N}dv(-) mutants were found to be altered in purine biosynthesis, strengthening the previous hypothesis of a symbiotic role of a bacterial purine derivative during the {N}od-independent symbiosis. {T}he other {N}dv(-) mutants were auxotrophic for pyrimidines and amino acids (leucine, glutamate, and lysine) or impaired in genes encoding proteins of unknown function. {T}he {F}ix(-) mutants were found to be affected in a wide variety of cellular processes, including both novel (n = 56) and previously identified (n = 31) genes important in symbiosis. {A}mong the novel genes identified, several were involved in the {C}alvin cycle, suggesting that {CO}2 fixation could play an important role during this symbiosis.}, keywords = {}, booktitle = {}, journal = {{M}olecular {P}lant-{M}icrobe {I}nteractions}, volume = {23}, numero = {6}, pages = {760--770}, ISSN = {0894-0282}, year = {2010}, DOI = {10.1094/mpmi-23-6-0760}, URL = {https://www.documentation.ird.fr/hor/fdi:010049531}, }
