@article{fdi:010049442, title = {{U}ltramafic soils from {N}ew {C}aledonia structure {P}isolithus albus in ecotype}, author = {{J}ourand, {P}hilippe and {D}ucousso, {M}. and {M}ajorel, {C}larisse and {H}annibal, {L}aure and {S}antoni, {S}. and {P}rin, {Y}. and {L}ebrun, {M}ichel}, editor = {}, language = {{ENG}}, abstract = {{I}solates of ectomycorrhizal {P}isolithus albus were sampled from both ultramafic and volcano-sedimentary soils in {N}ew {C}aledonia, a tropical hotspot of biodiversity, to investigate the relationships between genetic diversity and edaphic constraint through tolerance to nickel ({N}i). {C}arpophore description, spore morphology and phylogenetic analysis based on internal transcribed spacer ({ITS}) r{DNA} sequences confirmed that all isolates belong to {P}. albus and are closely related to other {A}ustralasian specimens. {U}sing molecular tools, {ITS}-restriction fragment length polymorphism and amplified fragment length polymorphism markers, we showed the existence of two distinct genetic clusters within {P}. albus: ultramafic and volcano-sedimentary. {M}ycelia response to {N}i toxicity supports such a population structure. {P}isolithus albus from ultramafic soils included isolates with a high diversity of in vitro {N}i tolerance, with both {N}i-tolerant isolates (average {N}i {EC}50 at 575 mu {M}) and {N}i-sensitive isolates (average {N}i {EC}50 at 37 mu {M}). {I}n contrast, all isolates from volcano-sedimentary soils were found to be {N}i sensitive (average {N}i {EC}50 at 32 mu {M}). {W}e highlight that (1) {P}. albus population from ultramafic soils of {N}ew {C}aledonia are genetically structured in ecotype, and that (2) {N}i tolerance among ultramafic isolates suggests an adaptive physiological response to {N}i toxicity.}, keywords = {{P}isolithus albus ; serpentine soils ; {N}i tolerance ; {N}ew {C}aledonia ; {NOUVELLE} {CALEDONIE}}, booktitle = {}, journal = {{F}ems {M}icrobiology {E}cology}, volume = {72}, numero = {2}, pages = {238--249}, ISSN = {0168-6496}, year = {2010}, DOI = {10.1111/j.1574-6941.2010.00843.x}, URL = {https://www.documentation.ird.fr/hor/fdi:010049442}, }
