@article{fdi:010048375, title = {{T}ransgenic coffee fruits from {C}offea arabica genetically modified by bombardment}, author = {{A}lbuquerque, {E}. {V}. {S}. and {C}unha, {W}. {G}. and {B}arbosa, {A}. {E}. {A}. {D}. and {C}osta, {P}. {M}. and {T}eixeira, {J}. {B}. and {V}ianna, {G}. {R}. and {C}abral, {G}. {B}. and {F}ernandez, {D}iana and {G}rossi-de-{S}a, {M}. {F}.}, editor = {}, language = {{ENG}}, abstract = {{T}he genetic modification of {C}offea arabica fruits is an important tool for the investigation of physiological characteristics and functional validation of genes related to coffee bean quality traits. {I}n this work, plants of {C}. arabica cultivar {C}atuai {V}ermelho were successfully genetically modified by bombardment of embryogenic calli. {C}alli were obtained from 90% of the leaf explants cultivated in a callogenesis-inducing medium modified with 20 mu {M} 2,4-dichlorophenoxyacetic acid (2,4-{D}). {T}he resulting calli were bombarded with the p{BI}426 vector containing a uid{A} and npt{II} gene fusion that was driven by the double {C}a{MV}35s promoter. {K}anamycin-selected embryos were positive for beta-glucuronidase ({GUS}) activity in histochemical assays and for target gene amplification by polymerase chain reaction. {I}ntegration of the npt{II} gene was confirmed by {S}outhern blot and showed a low copy number (one to three) of insertions. {T}ransformed plants showed normal development and settled fruits. {GUS} expression was assessed in the flower and fruit organs demonstrating the capacity of the double {C}a{MV}35s promoter to drive long-term stable expression of uid{A} in {C}. arabica fruit tissues. {M}oreover, we obtained a {T}-1 progeny presenting 3:1 {M}endelian segregation of the uid{A} gene. {T}his investigation is the first to report exogenous gene expression in coffee fruits and transgenic inheritance in {C}. arabica plants.}, keywords = {{C}offea arabica ; {G}enetic transformation ; uid{A} gene ; {C}offee fruit ; expression ; {B}ombardment}, booktitle = {}, journal = {{I}n {V}itro {C}ellular and {D}evelopmental {B}iology - {P}lant}, volume = {45}, numero = {5}, pages = {532--539}, ISSN = {1054-5476}, year = {2009}, DOI = {10.1007/s11627-009-9254-2}, URL = {https://www.documentation.ird.fr/hor/fdi:010048375}, }
