@article{fdi:010046260, title = {{P}rocessing of a dicistronic t{RNA}-sno{RNA} precursor : combined analysis in vitro and in vivo reveals alternate pathways and coupling to assembly of sno{RNP}}, author = {{B}arbezier, {N}. and {C}anino, {G}. and {R}odor, {J}. and {J}obet, {E}. and {S}aez-{V}asquez, {J}. and {M}archfelder, {A}. and {E}cheverria, {M}anuel}, editor = {}, language = {{ENG}}, abstract = {{T}he {C}/{D} box small nucleolar {RNA}s (sno{RNA}s) represent an essential class of small nucleolar {RNA}s that guide 2'-{O}-{R}ib methylation of ribosomal {RNA}s and other {RNA}s in eukaryotes. {I}n {A}rabidopsis ({A}rabidopsis thaliana), > 100 {C}/{D} sno{RNA}s have been identified, most of them encoded by polycistronic gene clusters, but little is known on the factors controlling their biogenesis. {H}ere, we focus on the identification of factors controlling the processing of t{RNA}-sno{RNA} dicistronic precursors (pre-tsno{RNA}) synthesized by {RNA} polymerase {III} and producing t{RNA}({G}ly) and {C}/{D} sno{R}43. {W}e produced radiolabeled {RNA} probes corresponding to different pre-tsno{RNA} mutants to test their impact on processing in vitro by a recombinant t{RNA}se {Z}, the {A}rabidopsis endonuclease that processes the 3' end of t{RNA}s, and by nuclear extracts from cauliflower ({B}rassica oleracea) inflorescences that accurately process the pre-tsno{RNA}. {T}his was coupled to an in vivo analysis of the processing of tagged pre-tsno{RNA} mutants expressed in {A}rabidopsis. {O}ur results strongly implicate t{RN}ase {Z} in endonucleolytic cleavage of the pre-tsno{RNA}. {I}n addition, they reveal an alternate pathway that could depend on a t{RNA} decay surveillance mechanism. {F}inally, we provide arguments showing that processing of pre-tsno{RNA}, both in planta and by nuclear extracts, is coupled to the assembly of sno{RNA} with core proteins forming the functional sno{RNP} (for small nucleolar ribonucleoprotein complex).}, keywords = {}, booktitle = {}, journal = {{P}lant {P}hysiology}, volume = {150}, numero = {3}, pages = {1598--1610}, ISSN = {0032-0889}, year = {2009}, DOI = {10.1104/pp.109.137968}, URL = {https://www.documentation.ird.fr/hor/fdi:010046260}, }
