@article{fdi:010044360, title = {{D}etection and quantification of root-knot nematode ({M}eloidogyne javanica), lesion nematode ({P}ratylenchus zeae) and dagger nematode ({X}iphinema elongatum) parasites of sugarcane using real-time {PCR}}, author = {{B}erry, {S}. {D}. and {F}argette, {M}ireille and {S}paull, {V}. {W}. and {M}orand, {S}. and {C}adet, {P}atrice}, editor = {}, language = {{ENG}}, abstract = {{A} number of different plant parasitic nematode species are found associated with sugarcane in {S}outh {A}frica. {O}f these, the root-knot nematode ({M}eloidogyne javanica), the lesion nematode ({P}ratylenchus zeae) and the dagger nematode ({X}iphinema elongatum) are potentially the most damaging pests. {I}dentification and enumeration of the number of these nematodes are necessary for providing advice to farmers as well as studying the effects of various treatments in field and glasshouse trials. {W}e report on the development, use, and extent of specificity of three sets of primers, for {M}. javanica, {P}. zeae and {X}. elongatum, and on tests to detect and quantify the number of these nematodes in soil samples using {SYBR} {G}reen {I} dye and real-time {PCR} technology. {A}mplicons from the three target species (obtained with their respective primer sets) are discernible in size by gel electrophoresis (380 bp for {M}. javanica, 250 bp for {P}. zeae and 500 bp for {X}. elongatum). {A}lso, these amplicons have cha! racteristic melting temperatures of 83.8 degrees {C} ({M}. javanica), 86.6 degrees {C} ({P}. zeae) and 86.1 degrees {C} ({X}. elongatum). {I}nvestigations into multiplex reactions found competition between species with {M}. javanica competing with {P}. zeae and {X}. elongatum. {S}ubsequent single tube (simplex) assays, enabled the construction of calibration curves for each of the three species. {T}hese were then used for quanti. cation of the numbers of each of these species in nematode samples extracted from the field, with a high ({R}-2 = 0.83) and significant positive correlation between real-time {PCR} and counts performed with microscopy.}, keywords = {{ITS}1 ; {M}icroscopy ; {P}lant parasitic nematodes ; {R}eal-time {PCR} ; {S}pecificity ; {S}ugarcane}, booktitle = {}, journal = {{M}olecular and {C}ellular {P}robes}, volume = {22}, numero = {3}, pages = {168--176}, ISSN = {0890-8508}, year = {2008}, DOI = {10.1016/j.mcp.2008.01.003}, URL = {https://www.documentation.ird.fr/hor/fdi:010044360}, }
