@article{fdi:010044053, title = {{B}iological and molecular characterization of a putative new sobemovirus infecting {I}mperata cylindrica and maize in {A}frica}, author = {{S}{\'e}r{\'e}m{\'e}, {D}. and {L}acombe, {S}{\'e}verine and {K}onat{\'e}, {M}. and {P}inel {G}alzi, {A}gn{\`e}s and {T}raor{\'e}, {V}. {S}. {E}. and {H}{\'e}brard, {E}ug{\'e}nie and {T}raor{\'e}, {O}. and {B}rugidou, {C}hristophe and {F}argette, {D}enis and {K}onat{\'e}, {G}.}, editor = {}, language = {{ENG}}, abstract = {{A} new virus was isolated from both the grass {I}mperata cylindrica and maize plants that had yellow mottle symptoms in {B}urkina {F}aso, {W}est {A}frica. {T}he virus has isometric particles ca. 32 nm in diameter. {T}he experimental host range was restricted to {R}ottboellia exaltata. {V}irions were isolated from leaves of systemically infected maize plants. {K}och's postulates were completed by mechanically inoculating uninfected {I}mperata or maize with either purified virus or sap from infected {I}mperata plants. {V}irion preparations were used to produce a specific polyclonal antiserum, and an enzyme-linked immunosorbent assay test was set up. {T}he full genome of the virus was sequenced, and it comprised 4,547 nucleotides. {P}hylogenetic studies indicated that the virus is closely related to rice yellow mottle virus, a sobemovirus that infects monocotyledons in {A}frica, and is more distantly related to cocksfoot mottle virus, another sobemovirus that infects monocotyledons. {A}lthough the virus can infect {R}. exaltata experimentally, it differs from {R}ottboellia yellow mottle virus, a member of a tentative species of the genus {S}obemovirus that also infects monocotyledons in {A}frica. {P}article morphology, serological properties, genomic organization, and phylogenetic analysis are all consistent with assignment of the new virus to the genus {S}obemovirus. {T}he name {I}mperata yellow mottle virus is proposed.}, keywords = {}, booktitle = {}, journal = {{A}rchives of {V}irology}, volume = {153}, numero = {10}, pages = {1813--1820}, ISSN = {0304-8608}, year = {2008}, DOI = {10.1007/s00705-008-0190-y}, URL = {https://www.documentation.ird.fr/hor/fdi:010044053}, }
