@article{fdi:010042562, title = {{P}ost-transcriptional gene silencing in the root system of the actinorhizal tree {A}llocasuarina verticillata}, author = {{G}herbi, {H}assen and {N}ambiar-{V}eetil, {M}athish and {Z}hong, {C}honglu and {F}{\'e}lix, {J}essy and {A}utran, {D}aphn{\'e} and {G}irardin, {R}apha{\¨e}l and {V}aissayre, {V}irginie and {A}uguy, {F}lorence and {B}ogusz, {D}idier and {F}ranche, {C}laudine}, editor = {}, language = {{ENG}}, abstract = {{I}n recent years, {RNA} interference has been exploited as a tool for investigating gene function in plants. {W}e tested the potential of double-stranded {RNA} interference technology for silencing a transgene in the actinorhizal tree {A}llocasuarina verticillata. {T}he approach was undertaken using stably transformed shoots expressing the beta-glueuronidase ({GUS}) gene under the control of the constitutive promoter 35{S}; the shoots were further transformed with the {A}grobacterium rhizogenes {A}4{RS} containing hairpin {RNA} (hp{RNA}) directed toward the {GUS} gene, and driven by the 35{S} promoter. {T}he silencing and control vectors contained the reporter gene of the green fluorescent protein ({GFP}), thus allowing a screening of {GUS}-silenced composite plantlets for autofluorescence. {W}ith this rapid procedure, histochemical data established that the reporter gene was strongly silenced in both fluorescent roots and actinorhizal nodules. {F}luorometric data further established that the level of {GUS} silencing was usually greater than 90% in the hairy roots containing the hairpin {GUS} sequences. {W}e found that the silencing process of the reporter gene did not spread to the aerial part of the composite {A}. verticillata plants. {R}eal-time quantitative polymerase chain reaction showed that {GUS} m{RNA}s were substantially reduced in roots and, thereby, confirmed the knock-down of the {GUS} transgene in the {GFP}(+) hairy roots. {T}he approach described here will provide a versatile tool for the rapid assessment of symbiotically related host genes in actinorhizal plants of the {C}asuarinaceae family.}, keywords = {}, booktitle = {}, journal = {{M}olecular {P}lant {M}icrobe {I}nteractions}, volume = {21}, numero = {5}, pages = {518--524}, ISSN = {0894-0282}, year = {2008}, DOI = {10.1094/{MPMI} -21-5-0518}, URL = {https://www.documentation.ird.fr/hor/fdi:010042562}, }
