@article{fdi:010040946, title = {{D}evelopment of genomic microsatellite markers in {C}offea canephora and their transferability to other coffee species}, author = {{P}oncet, {V}al{\'e}rie and {D}ufour, {M}. and {H}amon, {P}erla and {H}amon, {S}erge and {K}ochko, {A}lexandre de and {L}eroy, {T}.}, editor = {}, language = {{ENG}}, abstract = {{O}f the 103 accepted {C}offea species, 70% are threatened with extinction but only a few of them have been studied. {A} set of 40 polymorphic microsatellite markers was developed using a {GA}/{GT}-enriched {C}offea canephora genomic library. {A}mplification of these markers was tested in accessions of {C}. heterocalyx (a {C}ritically {E}ndangered species) and {C} pseudozanguebariae (a {V}ulnerable species) belonging to different {A}frican geographical clades. {A}ll microsatellites were polymorphic in {C}. canephora, with a mean allele number per polymorphic locus of more than 3 (at least 9 genotypes were tested). {O}bserved and expected heterozygosities calculated for {C}. canephora and {C}. pseudozanguebariae varied from 0.10 to 0.91 and from 0.20 to 0.77, respectively. {I}n total, 38 primer pairs (95%) were amplified in {C}. heterocalyx and {C}. pseudozanguebariae, indicating their high level of transferability across the genus {C}offea. {T}his large marker set will be useful for more extensive genetic studies of threatened {C}offea species.}, keywords = {}, booktitle = {}, journal = {{G}enome}, volume = {50}, numero = {12}, pages = {1156--1161}, ISSN = {0831-2796}, year = {2007}, DOI = {10.1139/{G}07-073}, URL = {https://www.documentation.ird.fr/hor/fdi:010040946}, }
