@article{fdi:010018010, title = {{A} phage single-stranded {DNA} (ss{DNA}) binding protein complements ss{DNA} accumulation of a geminivirus and interferes with viral movement}, author = {{P}adidam, {M}. and {B}eachy, {R}.{N}. and {F}auquet, {C}laude}, editor = {}, language = {{ENG}}, abstract = {{G}eminiviruses are plant viruses with circular single-stranded {DNA} (ss{DNA}) genomes encapsidated in double icosahedral particles. {T}omato leaf curl geminivirus ({T}o{LCV}) requires coat protein ({CP}) for the accumulation of ss{DNA} in protopolasts and in plants but not for systemic infection and symptom development in plants. {I}n the absence of {CP}, infected protoplasts accumulate reduced levels of ss{DNA} and increased amounts of double-stranded {DNA} (ds{DNA}), compared to accumulation in the presence of wild-type virus. {T}o determine whether the gene 5 protein (g5p), an ss{DNA} binding protein from #{E}scherichia coli$ phage {M}13, could restore the accumulation of ss{DNA}, {T}o{LCV} that lacked the {CP} gene was modified to express g5p or g5p fused to the {N}-terminal 66 amino acides of {CP} ({CP}66:6{G}:g5). {T}he modified viruses led to the accumulation of wild-type levels of ss{DNA}. {T}he high levels of ds{DNA} accumulation during infections with the modified viruses suggested a direct role for {CP} in viral {DNA} replication. {T}o{LCV} that produced the {CP}66:6{G}:g5 protein did not spread efficiently in #{N}icotiana benthamiana$ plants, and inoculated plants developed only very mild symptoms. {I}n infected protoplasts, the {CP}66:6{G}:g5 protein was immunolocalized to nuclei. {W}e propose that the fusion protein interferes with the function of the {BV}1 movement and thereby prevents spread of the infection.
