@article{fdi:010016015, title = {{M}olecular characterization of the incitant of cowpea bacterial blight and pustule, {X}anthomonas campestris pv. vignicola}, author = {{V}erdier, {V}al{\'e}rie and {A}ssigbets{\'e}, {K}omi and {K}hatri-{C}hhetri, {G}. and {W}ydra, {K}. and {R}udolph, {K}. and {G}eiger, {J}ean-{P}aul}, editor = {}, language = {{ENG}}, abstract = {{S}trains of #{X}anthomonas campestris$ pv. #vignicola$ ({X}cv), isolated from cowpea leaves with blight or minute pustules and collected from various geographic areas, were selected on the basis of pathological and physiological features. {A}ll strains were analyzed for genotypic markers by two methods : ribotyping with {E}co{RI} endonuclease, and {RFLP} analysis with a plasmid probe (pth{B}) containing a gene required for pathogenicity from #{X}anthomonas campestris$ pv. #manihotis$. {R}ibotyping revealed a unique pattern for all the strains that corresponded to the previously described ribotype r{RNA}7. {B}ased on polymorphism detected by pth{B} among {X}cv strains, nine haplotypes were defined. {T}he observed genetic variation was independent of the geographic origin of the strains and of pathogenic variation. {S}ome haplotypes were widely distributed, whereas others were localized. {I}n some cases, we could differentiate strains isolated from blight symptoms and pustules according to haplotypic composition. {H}owever, in most cases, no significant differences were observed. {O}ur results and the previous pathogenic and biochemical characterizations suggest that the strains isolated from leaves with blight symptoms or minute pustules belong to the same pathovar. {W}e provide information on pathogen diversity that can be used to identify and characterize resistant germplasm.
