Using three successive selections with plaque agarose overlay, 12 clones were established. Among them, five were selected for their better in vitro multiplication after being tested with a specific DNA probe and amplified both in vitro and in vivo. Restriction endonuclease analysis of the DNA extract demonstrated minor genotype differences between the clones, indicating the genetic heterogeneity of the viral population of the Tunisian isolate of PTMGV.
