@article{fdi:010008999, title = {{S}tudy of the dynamics of neutralization escape mutants in a chimpanzee naturally infected with the {S}imian {I}mmunodeficiency {V}irus {SIV}cpz-ant}, author = {{N}yambi, {P}.{N}. and {L}ewi, {P}. and {P}eeters, {M}artine and {J}anssens, {W}. and {H}eyndrickx, {L}. and {F}ransen, {K}. and {A}ndries, {K}. and {V}anden {H}aesevelde, {M}. and {H}eeney, {J}. and {P}iot, {P}. and {V}an {D}er {G}roen, {G}.}, editor = {}, language = {{ENG}}, abstract = {{H}ere we report on the use of spectral map analysis of time-paired sequential neutralization data of 11 serum samples of a chimpanzee naturally infected with a simian immunodeficiency virus ({SIV}cpz-ant) and 8 primary consecutive {SIV}cpz-ant isolates, taken at about 4-month intervals. {T}he analysis reveals the existence of three {SIV}cpz-ant isolate and serum neutralization clusters. {E}ach cluster group virus isolates and/or sera based on similarities of their neutralization spectra. {O}n average, neutralization escape mutants emerged after 15 months and mounted a neutralization response approximately 8 months later. {T}he entire gp160 regions of eight consecutive isolates were sequenced and analyzed by a new statistical method called polygram, which allowed the deduction of amino acid sequence motifs of gp160 which were specific for {SIV}cpz-ant isolates belonging to the same isolate neutralization clusters. {C}hanges in specific amino acid quadruplets in {V}1, {V}2, {C}3, {V}4, {V}5, and {CD}4 domains of gp120 and gp40 were seen to correlate with the neutralization clusters with most of the specific changes occuring in the {V}4 region. {T}his method of analysis may facilitate an understanding of the study of the dynamic interplay between human immunodeficiency virus ({HIV}) and host neutralization responses as well as providing possible insights into mechanisms of persistance of {HIV}-1 related lentiviruses in their natural hosts.
