@article{PAR00009521, title = {{A}ntibacterial, antifungal and antileishmanial activities of indolone-{N}-oxide derivatives}, author = {{I}brahim, {H}. and {F}uriga, {A}. and {N}ajahi, {E}. and {H}enocq, {C}. {P}. and {N}allet, {J}. {P}. and {R}oques, {C}. and {A}ubouy, {A}gn{\`e}s and {S}auvain, {M}ichel and {C}onstant, {P}. and {D}affe, {M}. and {N}epveu, {F}ran{\c{c}}oise}, editor = {}, language = {{ENG}}, abstract = {{A}n alarming increase in microbial resistance to traditional drugs and classical pharmacophores has spurred the search for new antimicrobial compounds. {I}ndolone-{N}-oxides ({INOD}s) possess a redox pharmacophore with promising, recently established, antimalarial activities. {I}n this study, the anti-infectious properties of a series of {INOD}s were investigated. {T}he antibacterial activity was evaluated against five bacterial strains {G}ram-positive ({S}taphylococcus aureus, {E}nterococcus hirae), {G}ram-negative ({P}seudomonas aeruginosa, {E}scherichia coli) and acid-fast ({M}ycobacterium tuberculosis). {T}he antifungal activity was assessed using two fungal strains ({A}spergillus niger, {C}andida albicans). {T}he antileishmanial activity was tested against two leishmanial strains, axenically-cultured amastigote ({L}eishmania infantum, {L}eishmania amazonensis). {T}he pharmacological activities are discussed as a function of structural and lipophilic characteristics. {T}he {G}ram-positive bacterial strain {E}. hirae was found to be the most sensitive strain, whereas the {G}ram-negative {E}. coli was resistant to this family of compounds. {O}ne compound (64) was more potent than nalidixic acid against {E}. hirae, whereas another one (52) was equipotent as clotrimazole against {C}. albicans. {INOD}s were microbe -cidal rather than -static. {INOD}s showed good antitubercular activity in the low micromolar range (similar to ciprofloxacin). {I}n addition, {INOD}-antiprotozoal potencies were confirmed against the leishmania parasite. {INOD}s showed a broad spectrum of antimicrobial activity and offer a promising anti-infectious prototype worthy of being developed.}, keywords = {antibacterial ; antifungal ; antileishmanial ; antitubercular ; broad spectrum ; indolone-{N}-oxide ; redox pharmacophore}, booktitle = {}, journal = {{J}ournal of {A}ntibiotics}, volume = {65}, numero = {10}, pages = {499--504}, ISSN = {0021-8820}, year = {2012}, DOI = {10.1038/ja.2012.60}, URL = {https://www.documentation.ird.fr/hor/{PAR}00009521}, }
