@article{PAR00007807, title = {{F}irst evidence for a direct inhibitory effect of kisspeptins on {LH} expression in the eel, {A}nguilla anguilla}, author = {{P}asquier, {J}. and {L}afont, {A}. {G}. and {L}eprince, {J}. and {V}audry, {H}. and {R}ousseau, {K}. and {D}ufour, {S}ylvie}, editor = {}, language = {{ENG}}, abstract = {{T}he kisspeptin system has emerged as one of the main puberty gatekeepers among vertebrates. {T}he {E}uropean eel ({A}nguilla anguilla) is a remarkable model due to its phylogenetical position at the basis of teleosts, and its unique life cycle with a blockade of puberty before reproductive migration. {W}e cloned the full-length coding sequence of a kisspeptin receptor ({K}issr) in the eel. {C}omparison of {K}issr sequences assigned the eel {K}issr to a basal position in a clack including most of the known teleost {K}issr, in agreement with the eel phylogenetical position. {E}el {K}issr tissue distribution was analyzed by quantitative real-time {PCR}. {E}el {K}issr was highly expressed in the brain, especially in the telencephalon and di-/mesencephalon, while a very low or undetectable expression was observed in various peripheral organs. {A} high expression of {K}issr was also found in the pituitary indicating a possible direct pituitary role of kisspeptin. {P}rimary cultures of eel pituitary cells were performed to investigate the direct effects of kisspeptin on pituitary hormone expression. {H}uman/lamprey kisspeptin exerted a time- and dose-dependent inhibitory effect on {LH} beta expression. {A}ll other tested kisspeptins had a similar inhibitory effect on {LH} beta expression. {T}he inhibitory effect of kisspeptins was exerted specifically on {LH} beta as no change was induced on the expression of other glycoprotein hormone subunits ({GP} alpha, {FSH} beta and {TSH} beta) nor of growth hormone. {T}hese data provide the first evidence for the existence. in the {E}uropean eel, of a kisspeptin system, which may play a direct inhibitory role on pituitary {LH} beta expression.}, keywords = {{K}isspeptin receptor ; {P}ituitary cells ; {G}onadotropin ; {T}eleost ; {E}uropean ; eel}, booktitle = {}, journal = {{G}eneral and {C}omparative {E}ndocrinology}, volume = {173}, numero = {1}, pages = {216--225}, ISSN = {0016-6480}, year = {2011}, DOI = {10.1016/j.ygcen.2011.05.019}, URL = {https://www.documentation.ird.fr/hor/{PAR}00007807}, }
