@article{PAR00006598, title = {{A}t{NUFIP}, an essential protein for plant development, reveals the impact of sno{RNA} gene organisation on the assembly of sno{RNP}s and r{RNA} methylation in {A}rabidopsis thaliana}, author = {{R}odor, {J}. and {J}obet, {E}. and {B}izarro, {J}. and {V}ignols, {F}. and {C}arles, {C}. and {S}uzuki, {T}. and {N}akamura, {K}. and {E}cheverria, {M}anuel}, editor = {}, language = {{ENG}}, abstract = {{P}>{I}n all eukaryotes, {C}/{D} small nucleolar ribonucleoproteins ({C}/{D} sno{RNP}s) are essential for methylation and processing of ribosomal {RNA}s. {T}hey consist of a box {C}/{D} small nucleolar {RNA} ({C}/{D} sno{RNA}) associated with four highly conserved nucleolar proteins. {R}ecent data in {H}e{L}a cells and yeast have revealed that assembly of these sno{RNP}s is directed by {NUFIP} protein and other auxiliary factors. {N}evertheless, the precise function and biological importance of {NUFIP} and the other assembly factors remains unknown. {I}n plants, few studies have focused on {RNA} methylation and sno{RNP} biogenesis. {H}ere, we identify and characterise the {A}t{NUFIP} gene that directs assembly of {C}/{D} sno{RNP}. {T}o elucidate the function of {A}t{NUFIP} in planta, we characterized atnufip mutants. {T}hese mutants are viable but have severe developmental phenotypes. {N}orthern blot analysis of sno{RNA} accumulation in atnufip mutants revealed a specific degradation of {C}/{D} sno{RNA}s and this situation is correlated with a reduction in r{RNA} methylation. {R}emarkably, the impact of {A}t{NUFIP} depends on the structure of sno{RNA} genes: it is essential for the accumulation of those {C}/{D} sno{RNA}s encoded by polycistronic genes, but not by monocistronic or tsno{RNA} genes. {W}e propose that {A}t{NUFIP} controls the kinetics of {C}/{D} sno{RNP} assembly on nascent precursors to overcome sno{RNA} degradation of aberrant {RNP}s. {F}inally, we show that {A}t{NUFIP} has broader {RNP} targets, controlling the accumulation of sca{RNA}s that direct methylation of spliceosomal sn{RNA} in {C}ajal bodies.}, keywords = {{A}t{NUFIP} ; sno{RNP} assembly ; atnuf mutants ; development ; r{RNA} methylation}, booktitle = {}, journal = {{P}lant {J}ournal}, volume = {65}, numero = {5}, pages = {807--819}, ISSN = {0960-7412}, year = {2011}, DOI = {10.1111/j.1365-313{X}.2010.04468.x}, URL = {https://www.documentation.ird.fr/hor/{PAR}00006598}, }
