@article{PAR00004635, title = {{A}pplicability of the 16{S}-23{S} r{DNA} internal spacer for {PCR} detection of the phytostimulatory {PGPR} inoculant {A}zospirillum lipoferum {CRT}1 in field soil}, author = {{B}audoin, {E}z{\'e}kiel and {C}ouillerot, {O}. and {S}paepen, {S}. and {M}oenne-{L}occoz, {Y}. and {N}azaret, {S}.}, editor = {}, language = {{ENG}}, abstract = {{A}ims: {T}o assess the applicability of the 16{S}-23{S} r{DNA} internal spacer regions ({ISR}) as targets for {PCR} detection of {A}zospirillum ssp. and the phytostimulatory plant growth-promoting rhizobacteria seed inoculant {A}zospirillum lipoferum {CRT}1 in soil. {M}ethods and {R}esults: {P}rimer sets were designed after sequence analysis of the {ISR} of {A}. lipoferum {CRT}1 and {A}zospirillum brasilense {S}p245. {T}he primers f{AZO}/r{AZO} targeting the {A}zospirillum genus successfully yielded {PCR} amplicons (400-550 bp) from {A}zospirillum strains but also from certain non-{A}zospirillum strains in vitro, therefore they were not appropriate to monitor indigenous {A}zospirillum soil populations. {T}he primers f{CRT}1/r{CRT}1 targeting {A}. lipoferum {CRT}1 generated a single 249-bp {PCR} product but could also amplify other strains from the same species. {H}owever, with {DNA} extracts from the rhizosphere of field-grown maize, both f{AZO}/r{AZO} and f{CRT}1/r{CRT}1 primer sets could be used to evidence strain {CRT}1 in inoculated plants by nested {PCR}, after a first {ISR} amplification with universal ribosomal primers. {I}n soil, a 7-log dynamic range of detection (102-108 {CFU} g-1 soil) was obtained. {C}onclusions: {T}he {PCR} primers targeting 16{S}-23{S} r{DNA} {ISR} sequences enabled detection of the inoculant {A}. lipoferum {CRT}1 in field soil. {S}ignificance and {I}mpact of the {S}tudy: {C}onvenient methods to monitor {A}zospirillum phytostimulators in the soil are lacking. {T}he {PCR} protocols designed based on {ISR} sequences will be useful for detection of the crop inoculant {A}. lipoferum {CRT}1 under field conditions.}, keywords = {{A}zospirillum ; plant growth-promoting rhizobacteria ; specific primers ; soil detection ; 16{S}-23{S} r{DNA} intergenic spacer}, booktitle = {}, journal = {{J}ournal of {A}pplied {M}icrobiology}, volume = {108}, numero = {1}, pages = {25--38}, ISSN = {1364-5072}, year = {2010}, DOI = {10.1111/j.1365-2672.2009.04393.x}, URL = {https://www.documentation.ird.fr/hor/{PAR}00004635}, }
