@article{PAR00002538, title = {{P}otentiation of {NK} cell-mediated cytotoxicity in human lung adenocarcinoma : role of {NKG}2{D}-dependent pathway}, author = {{L}e {M}aux {C}hansac, {B}. and {M}iss{\'e}, {D}oroth{\'e}e and {R}ichon, {C}. and {V}ergnon, {I}. and {K}ubin, {M}. and {S}oria, {J}. {C}. and {M}oretta, {A}. and {C}houaib, {S}. and {M}ami-{C}houaib, {F}.}, editor = {}, language = {{ENG}}, abstract = {{N}atural cytotoxicity receptors and {NKG}2{D} correspond to major activating receptors involved in triggering of tumor cell lysis by human {NK} cells. {I}n this report, we investigated the expression of {NKG}2{D} ligands ({NKG}2{DL}s), {MHC} class {I}-related chain ({MIC}) {A}, {MICB} and {UL}16-binding proteins 1, 2 and 3, on a panel of human non-small-cell lung carcinoma cell lines, and we analyzed their role in tumor cell susceptibility to {NK} cell lysis. {A}lthough adenocarcinoma ({ADC}) cells expressed heterogeneous levels of {NKG}2{DL}s, they were often resistant to {NK} cell-mediated killing. {R}esistance of a selected cell line, {ADC}-{C}oco, to allogeneic polyclonal {NK} cells and autologous {NK} cell clones correlated with shedding of {NKG}2{DL}s resulting from a matrix metalloproteinase ({MMP}) production. {T}reatment of {ADC}-{C}oco cells with a {MMP} inhibitor ({MMPI}) combined with {IL}-15 stimulation of autologous {NK} cell clones lead to a potentiation of {NK} cell-mediated cytotoxicity. {T}his lysis is mainly {NKG}2{D} mediated, since it is abrogated by anti-{NKG}2{D}-neutralizing m{A}b. {T}hese results suggest that {MMPI}s, in combination with {IL}-15, may be useful for overcoming tumor cell escape from the innate immune response.}, keywords = {{N}k cell-activating receptors ; {N}kg2dl ; {N}sclc ; {T}umor-infiltrating nk cells}, booktitle = {}, journal = {{I}nternational {I}mmunology}, volume = {20}, numero = {7}, pages = {801--810}, ISSN = {0953-8178}, year = {2008}, DOI = {10.1093/intimm/dxn038}, URL = {https://www.documentation.ird.fr/hor/{PAR}00002538}, }
