@article{fdi:010088158, title = {{T}hermospira aquatica gen. nov., sp. nov., a novel thermophilic spirochete isolated from a {T}unisian hot spring, and description of the novel family {T}hermospiraceae}, author = {{B}en ali gam, {Z}ouhaier and {T}hioye, {A}. and {C}ayol, {J}. {L}. and {P}ostec, {A}. and {B}artoli-{J}oseph, {M}. and {V}andecasteele, {C}. and {E}rauso, {G}. and {L}abat, {M}arc}, editor = {}, language = {{ENG}}, abstract = {{A} novel thermophilic, anaerobic bacterium, strain {F}1{F}22{T}, was isolated from hot spring water collected in northern {T}unisia. {T}he cells were non-motile, {G}ram-negative and helical with hooked ends, 0.5x10-32 mu m in size. {G}rowth of the strain was observed at 45-70 degrees {C} (optimum, 55 degrees {C}), in 0.0-1.0 % (w/v) {N}a{C}l (optimum without {N}a{C}l) and at p{H} 6.5-8.5 (optimum, p{H} 7.5). {Y}east extract was required for growth, and the strain grew on glucose, sucrose and maltose. {T}he major fatty acids were {C}16:0 (40.2 %), iso- {C}16: 0 (30.2 %) and {C}16 :0 {DMA} (14.5 %). {T}he genome consisted of a circular chromosome (2.5 {M}b) containing 2672 predicted protein -encoding genes with a {G}+{C} content of 43.15 mol %. {B}ased on a comparative 16{S} r{RNA} gene sequence analysis, strain {F}1{F}22{T} formed a deeply branching lineage within the phylum {S}pirochaetota, class {S}pirochaetia, order {B}revinematales, and had only low sequence similarity to other species of the phylum (lower than 83 %). {G}enome- based analysis of average nucleotide identity and digital {DNA}-{DNA} hybridization of strain {F}1{F}22{T} with {T}reponema caldarium {DSM} 7334{T}, {B}revinema andersonii {ATCC} 43811{T} and {S}pirochaeta thermophila {DSM} 6578{T} showed values between 63.26 and 63.52 %, and between 20 and 25 %. {H}ence, we propose strain {F}1{F}22{T} as a representative of a novel family ({T}hermospiraceae fam. nov.), genus and species of {B}revinematales: {T}her-mospira aquatica gen. nov., sp. nov.
