@article{fdi:010086481, title = {{MALDI}-{TOF} {MS} limits for the identification of {M}editerranean sandflies of the subgenus {L}arroussius, with a special focus on the {P}hlebotomus perniciosus {C}omplex}, author = {{H}uguenin, {A}. and {P}esson, {B}. and {K}altenbach, {M}. {L}. and {D}iarra, {A}dama {Z}an and {P}arola, {P}. and {D}epaquit, {J}. {M}. and {R}andrianambinintsoa, {F}. {J}.}, editor = {}, language = {{ENG}}, abstract = {{L}eishmania infantum is the agent of visceral leishmaniasis in the {M}editerranean basin. {I}t is transmitted by sandflies of the subgenus {L}arroussius. {A}lthough {P}hlebotomus perniciosus is the most important vector in this area, an atypical {P}h. perniciosus easily confused with {P}h. longicuspis has been observed in {N}orth {A}frica. {MALDI}-{TOF} {MS}, an important tool for vector identification, has recently been applied for the identification of sandflies. {S}pectral databases presented in the literature, however, include only a limited number of {L}arroussius species. {O}ur objective was to create an in-house database to identify {M}editerranean sandflies and to evaluate the ability of {MALDI}-{TOF} {MS} to discriminate close species or atypical forms within the {L}arroussius subgenus. {F}ield-caught specimens (n = 94) were identified morphologically as typical {P}h. perniciosus ({PN}; n = 55), atypical {P}h. perniciosus ({PNA}; n = 9), {P}h. longicuspis (n = 9), {P}h. ariasi (n = 9), {P}h. mascittii (n = 3), {P}h. neglectus (n = 5), {P}h. perfiliewi (n = 1), {P}h. similis (n = 9) and {P}h. papatasi (n = 2). {I}dentifications were confirmed by sequencing of the mt{DNA} {C}yt{B} region and sixteen specimens were included in the in-house database. {B}lind assessment on 73 specimens (representing 1073 good quality spectra) showed a good agreement (98.5%) between {MALDI}-{TOF} {MS} and molecular identification. {D}iscrepancies concerned confusions between {P}h. perfiliewi and {P}h. perniciosus. {H}ierarchical clustering did not allow classification of {PN} and {PNA}. {T}he use of machine learning, however, allowed discernment between {PN} and {PNA} and between the lcus and lcx haplotypes of {P}h. longicuspis (accuracy: 0.8938 with partial-least-square regression and random forest models). {MALDI}-{TOF} {MS} is a promising tool for the rapid and accurate identification of field-caught sandflies. {T}he use of machine learning could allow to discriminate similar species.}, keywords = {{P}hlebotomus ; {L}arroussius ; species complex ; {MALDI}-{TOF} {MS} ; identification ; high-throughput identification ; molecular biology ; {PORTUGAL} ; {ESPAGNE} ; {FRANCE} ; {ITALIE} ; {MALTE} ; {MAROC} ; {PAYS} {MEDITERRANEENS}}, booktitle = {}, journal = {{M}icroorganisms}, volume = {10}, numero = {11}, pages = {2135 [14 ]}, year = {2022}, DOI = {10.3390/microorganisms10112135}, URL = {https://www.documentation.ird.fr/hor/fdi:010086481}, }
