@article{fdi:010082229, title = {{A}lkalicella caledoniensis gen. nov., sp. nov., a novel alkaliphilic anaerobic bacterium isolated from '{L}a {C}rouen' alkaline thermal spring, {N}ew {C}aledonia}, author = {{Q}u{\'e}m{\'e}neur, {M}arianne and {E}rauso, {G}. and {B}artoli, {M}. and {V}andecasteele, {C}. and {W}ils, {L}. and {G}il, {L}. and {M}onnin, {C}. and {P}elletier, {B}ernard and {P}ostec, {A}.}, editor = {}, language = {{ENG}}, abstract = {{A} novel anaerobic, alkaliphilic, mesophilic, {G}ram-stain-positive, endospore-forming bacterium was isolated from an alkaline thermal spring (42 degrees {C}, p{H} 9.0) in {N}ew {C}aledonia. {T}his bacterium, designated strain {LB}2({T}), grew at 25-50 degrees {C} (optimum, 37 degrees {C}) and p{H} 8.2-10.8 (optimum, p{H} 9.5). {A}dded {N}a{C}l was not required for growth (optimum, 0-1 %) but was tolerated up to 7 %. {S}train {LB}2({T}) utilized a limited range of substrates, such as peptone, pyruvate, yeast extract and xylose. {E}nd products detected from pyruvate fermentation were acetate and formate. {B}oth ferric citrate and thiosulfate were used as electron acceptors. {E}lemental sulphur, nitrate, nitrite, fumarate, sulphate, sulfite and {DMSO} were not used as terminal electron acceptors. {T}he two major cellular fatty acids were iso-{C}-15:0 and {C}-16:0. {T}he genome consists of a circular chromosome (3.7 {M}b) containing 3626 predicted protein-encoding genes with a {G}+{C} content of 36.2 mol%. {P}hylogenetic analysis based on the 16{S} r{RNA} gene sequence indicated that the isolate is a member of the family {P}roteinivoraceae, order {C}lostridiales within the phylum {F}irmicutes. {S}train {LB}2({T}) was most closely related to the thermophilic {A}naerobranca gottschalkii {LBS}3({T}) (93.2 % 16{S} r{RNA} gene sequence identity). {G}enome-based analysis of average nucleotide identity and digital {DNA}-{DNA} hybridization of strain {LB}2{T} with {A}. gottschalkii {LBS}3({T}) showed respective values of 70.8 and 13.4 %. {B}ased on phylogenetic, genomic, chemotaxonomic and physiological prop-erties, strain {LB}2({T}) is proposed to represent the first species of a novel genus, for which the name {A}lkalicella caledoniensis gen. nov., sp. nov. is proposed (type strain {LB}2({T})={DSM} 100588({T})={JCM} 30958({T})).}, keywords = {{F}irmicutes ; anaerobic ; alkaliphilic ; alkaline thermal spring ; {P}roteinivoraceae ; {A}lkalicella caledoniensis ; {NOUVELLE} {CALEDONIE}}, booktitle = {}, journal = {{I}nternational {J}ournal of {S}ystematic and {E}volutionary {M}icrobiology}, volume = {71}, numero = {5}, pages = {004810 [9 ]}, ISSN = {1466-5026}, year = {2021}, DOI = {10.1099/ijsem.0.004810}, URL = {https://www.documentation.ird.fr/hor/fdi:010082229}, }
