@article{fdi:010075622, title = {{SAMHD}1 enhances chikungunya and zika virus replication in human skin fibroblasts}, author = {{W}ichit, {S}. and {H}amel, {R}odolphe and {Z}anzoni, {A}. and {D}iop, {F}. and {C}ribier, {A}. and {T}alignani, {L}. and {D}iack, {A}. and {F}erraris, {P}auline and {L}i{\'e}geois, {F}lorian and {U}rbach, {S}. and {E}kchariyawat, {P}. and {M}erits, {A}. and {Y}ssel, {H}. and {B}enkirane, {M}. and {M}iss{\'e}, {D}oroth{\'e}e}, editor = {}, language = {{ENG}}, abstract = {{C}hikungunya virus ({CHIKV}) and {Z}ika virus ({ZIKV}) are emerging arboviruses that pose a worldwide threat to human health. {C}urrently, neither vaccine nor antiviral treatment to control their infections is available. {A}s the skin is a major viral entry site for arboviruses in the human host, we determined the global proteomic profile of {CHIKV} and {ZIKV} infections in human skin fibroblasts using {S}table {I}sotope {L}abelling by {A}mino acids in {C}ell culture ({SILAC})-based mass-spectrometry analysis. {W}e show that the expression of the interferon-stimulated proteins {MX}1, {IFIT}1, {IFIT}3 and {ISG}15, as well as expression of defense response proteins {DDX}58, {STAT}1, {OAS}3, {EIF}2{AK}2 and {SAMHD}1 was significantly up-regulated in these cells upon infection with either virus. {E}xogenous expression of {IFIT}s proteins markedly inhibited {CHIKV} and {ZIKV} replication which, accordingly, was restored following the abrogation of {IFIT}1 or {IFIT}3. {O}verexpression of {SAMHD}1 in cutaneous cells, or pretreatment of cells with the virus-like particles containing {SAMHD}1 restriction factor {V}px, resulted in a strong increase or inhibition, respectively, of both {CHIKV} and {ZIKV} replication. {M}oreover, silencing of {SAMHD}1 by specific {SAMHD}1-si{RNA} resulted in a marked decrease of viral {RNA} levels. {T}ogether, these results suggest that {IFIT}s are involved in the restriction of replication of {CHIKV} and {ZIKV} and provide, as yet unreported, evidence for a proviral role of {SAMHD}1 in arbovirus infection of human skin cells.}, keywords = {{SAMHD}1 ; arbovirus ; {C}hikungunya ; {Z}ika ; {IFIT} ; {SILAC}}, booktitle = {}, journal = {{I}nternational {J}ournal of {M}olecular {S}ciences}, volume = {20}, numero = {7}, pages = {art. 1695 [18 ]}, ISSN = {1422-0067}, year = {2019}, DOI = {10.3390/ijms20071695}, URL = {https://www.documentation.ird.fr/hor/fdi:010075622}, }
