@article{fdi:010073676, title = {{T}wo ancestral genes shaped the {X}anthomonas campestris {TAL} effector gene repertoire}, author = {{D}enance, {N}. and {S}zurek, {B}oris and {D}oyle, {E}. {L}. and {L}auber, {E}. and {F}ontaine-{B}odin, {L}. and {C}arrere, {S}. and {G}uy, {E}. and {H}ajri, {A}. and {C}erutti, {A}. and {B}oureau, {T}. and {P}oussier, {S}. and {A}rlat, {M}. and {B}ogdanove, {A}. {J}. and {N}oel, {L}. {D}.}, editor = {}, language = {{ENG}}, abstract = {{X}anthomonas transcription activator-like effectors ({TALE}s) are injected inside plant cells to promote host susceptibility by enhancing transcription of host susceptibility genes. {TALE}-encoding (tal) genes were thought to be absent from {B}rassicaceae-infecting {X}anthomonas campestris ({X}c) genomes based on four reference genomic sequences. {W}e discovered tal genes in 26 of 49 {X}c strains isolated worldwide and used a combination of single molecule real time ({SMRT}) and tal amplicon sequencing to yield a near-complete description of the {TALE}s found in {X}c ({X}c {TAL}ome). {T}he 53 sequenced tal genes encode 21 distinct {DNA} binding domains that sort into seven major {DNA} binding specificities. {I}n silico analysis of the {B}rassica rapa promoterome identified a repertoire of predicted {TALE} targets, five of which were experimentally validated using quantitative reverse transcription polymerase chain reaction. {T}he {X}c {TAL}ome shows multiple signs of {DNA} rearrangements that probably drove its evolution from two ancestral tal genes. {W}e discovered that {T}al12a and {T}al15a of {X}cc strain {X}ca5 contribute together in the development of disease symptoms on susceptible {B}.oleracea var. botrytis cv {C}lovis. {T}his large and polymorphic repertoire of {TALE}s opens novel perspectives for elucidating {TALE}-mediated susceptibility of {B}rassicaceae to black rot disease and for understanding the molecular processes underlying {TALE} evolution.}, keywords = {black rot ; {B}rassica rapa ; {B}rassicaceae ; {H}ax ; {TALE} ; {X}anthomonas campestris}, booktitle = {}, journal = {{N}ew {P}hytologist}, volume = {219}, numero = {1}, pages = {391--407}, ISSN = {0028-646{X}}, year = {2018}, DOI = {10.1111/nph.15148}, URL = {https://www.documentation.ird.fr/hor/fdi:010073676}, }
