@article{fdi:010073196, title = {{P}harmacological activity of {C}urarea toxicofera in combination with classical antimalarial treatments}, author = {{R}odriguez, {Y}. {V}. and {A}rias, {M}. {H}. and {G}arcia, {J}. {O}. and {D}eharo, {E}ric and {G}aravito, {G}.}, editor = {}, language = {{ENG}}, abstract = {{E}thnopharmacological relevance: {I}n the {L}eticia-{A}mazonas area, {U}itoto indigenous people use a preparation of {C}urarea toxicofera ({W}edd) {B}arneby & {K}rukoff ({M}enispermaceae) alone or combined with prescribed medications to prevent and treat malaria. {A}im of study: {T}o determine the in vitro and in vivo antiplasmodial activity of traditional preparations of {C}urarea toxicofera alone and in combination with classical antimalarials. {M}aterial and {M}ethods: {T}he traditional preparation was evaluated in vitro against {P}. falciparum {FCR}3 {CQ} resistant strain, alone and combined. {T}he preparation was further administered orally alone or combined with chloroquine and artesunate in mice infected with {P}lasmodium berghei {ANKA} strain on the four-day antimalarial test model. {R}esults: {T}he herbal remedy used alone was able to significantly decrease the parasitemia both in vitro ({IC}50 7.3 mu g/ml) and in vivo ({ED}50 328 mg/{K}g) but it was less active than chloroquine ({IC}50 0.29 mu g/ml in vitro and {ED}50 2.3 mg/{K}g/day in vivo), and than artesunate ({IC}50 0.002 mu g/ml and {ED}50 3.7 mg/{K}g/day). {I}nterestingly it presented synergism with chloroquine in vitro ({C}ombination {I}ndex: 0.39) and in vivo; and was additive with artesunate in vitro ({C}ombination {I}ndex: 0.94) and in vivo. {C}onclusion: {T}he traditional preparation showed potential as an antimalarial and, when used in combination, does not negatively affect the efficacy of the drugs evaluated. {P}re-clinical studies should be conducted with a standardized preparation to confirm its efficacy and safety alone and in combination with chloroquine and artesunate.}, keywords = {{A}ntimalarials ; {E}thnopharmacology ; {D}rug {I}nteractions ; {P}lasmodium ; {C}urarea toxicofera ; {COLOMBIE} ; {AMAZONIE}}, booktitle = {}, journal = {{J}ournal of {E}thnopharmacology}, volume = {222}, numero = {}, pages = {288--294}, ISSN = {0378-8741}, year = {2018}, DOI = {10.1016/j.jep.2018.04.008}, URL = {https://www.documentation.ird.fr/hor/fdi:010073196}, }
