@article{fdi:010073180, title = {{I}dentification and characterization of host proteins bound to dengue virus 3' {UTR} reveal an antiviral role for quaking proteins}, author = {{L}iao, {K}. {C}. and {C}huo, {V}. and {N}g, {W}. {C}. and {N}eo, {S}. {P}. and {P}ompon, {J}ulien and {G}unaratne, {J}. and {O}oi, {E}. {E}. and {G}arcia-{B}lanco, {M}. {A}.}, editor = {}, language = {{ENG}}, abstract = {{T}he four dengue viruses ({DENV}1-4) are rapidly reemerging infectious {RNA} viruses. {T}hese positive-strand viral genomes contain structured 3' untranslated regions ({UTR}s) that interact with various host {RNA} binding proteins ({RBP}s). {T}hese {RBP}s are functionally important in viral replication, pathogenesis, and defense against host immune mechanisms. {H}ere, we combined {RNA} chromatography and quantitative mass spectrometry to identify proteins interacting with {DENV}1-4 3' {UTR}s. {A}s expected, {RBP}s displayed distinct binding specificity. {A}mong them, we focused on quaking ({QKI}) because of its preference for the {DENV}4 3' {UTR} ({DENV}-4/{SG}/06{K}2270{DK}1/2005). {RNA} immunoprecipitation experiments demonstrated that {QKI} interacted with {DENV}4 genomes in infected cells. {M}oreover, {QKI} depletion enhanced infectious particle production of {DENV}4. {O}n the contrary, {QKI} did not interact with {DENV}2 3' {UTR}, and {DENV}2 replication was not affected consistently by {QKI} depletion. {N}ext, we mapped the {QKI} interaction site and identified a {QKI} response element ({QRE}) in {DENV}4 3' {UTR}. {I}nterestingly, removal of {QRE} from {DENV}4 3' {UTR} abolished this interaction and increased {DENV}4 viral particle production. {I}ntroduction of the {QRE} to {DENV}2 3' {UTR} led to {QKI} binding and reduced {DENV}2 infectious particle production. {F}inally, reporter assays suggest that {QKI} reduced translation efficiency of viral {RNA}. {O}ur work describes a novel function of {QKI} in restricting viral replication.}, keywords = {{QKI} ; dengue virus ; translation ; host factors ; {RNA} elements}, booktitle = {}, journal = {{RNA}}, volume = {24}, numero = {6}, pages = {803--814}, ISSN = {1355-8382}, year = {2018}, DOI = {10.1261/rna.064006.117}, URL = {https://www.documentation.ird.fr/hor/fdi:010073180}, }
