@article{fdi:010072417, title = {{C}ryopreservation of arecanut ({A}reca catechu l.) pollen}, author = {{K}arun, {A}. and {S}ajini, {K}. {K}. and {M}uralikrishna, {K}. {S}. and {R}ajesh, {M}. {K}. and {E}ngelmann, {F}lorent}, editor = {}, language = {{ENG}}, abstract = {{BACKGROUND}: {C}ryopreservation opens new avenues in the field of genetic resource conservation, especially in recalcitrant seeded palms such as arecanut for which field genebanks are exposed to pest and disease attacks and natural calamities. {I}t is only through cryopreservation that the safety of the conserved germplasm can be assured at a relatively low cost for extended periods. {OBJECTIVE}: {T}he objective of this work was to standardize various aspects of arecanut pollen cryopreservation, viz. collection and desiccation of pollen, in vitro germination, viability and fecundity studies. {MATERIALS} {AND} {METHODS}: {P}ollens of three arecanut genotypes ({S}umangala, {H}irehalli {D}warf and {H}irehalli {D}warf x {S}umangala) were collected in {D}ecember 2013-{F}ebruary 2014. {I}n vitro viability tests were conducted using fresh and desiccated pollen. {D}esiccated pollen was cryopreserved by direct immersion in liquid nitrogen and cryostored for different durations (24 hours to 2 years). {V}iability and fertility studies were conducted using cryopreserved pollen. {RESULTS}: {P}ollen extraction was achieved from fully opened male flowers by desiccation at room temperature (33-34 degrees {C}). {A} medium containing 2.5 g/{L} sucrose was found to be best for in vitro germination at room temperature. {T}here was no significant difference in germination between desiccated and cryopreserved pollen whereas pollen tube length decreased significantly after cryopreservation. {F}ertility studies using {HD} x {S}umangala pollen cryostored for various durations (1 month, 1 year and 2 years) showed the setting of 70, 43 and 62%, respectively. {N}ormal nut set was observed using cryopreserved pollen. {CONCLUSION}: {P}ollen cryopreservation is a viable option for germplasm conservation and hybridization programmes in arecanut.}, keywords = {{A}reca catechu {L}. ; arecanut ; pollen ; cryopreservation ; germination ; vigor ; fertility}, booktitle = {}, journal = {{C}ryoletters}, volume = {38}, numero = {6}, pages = {463--470}, ISSN = {0143-2044}, year = {2017}, URL = {https://www.documentation.ird.fr/hor/fdi:010072417}, }
