@article{fdi:010071510, title = {{C}haracterization of recombinant {T}rypanosoma brucei gambiense {T}ranslationally {C}ontrolled {T}umor {P}rotein (r{T}bg{TCTP}) and its interaction with {G}lossina bacteria}, author = {{B}ossard, {G}. and {B}artoli, {M}. and {F}ardeau, {M}arie-{L}aure and {H}olzmuller, {P}. and {O}llivier, {B}ernard and {G}eiger, {A}nne}, editor = {}, language = {{ENG}}, abstract = {{I}n humans, sleeping sickness (i.e. {H}uman {A}frican {T}rypanosomiasis) is caused by the protozoan parasites {T}rypanosoma brucei gambiense ({T}bg) in {W}est and {C}entral {A}frica, and {T}. b. rhodesiense in {E}ast {A}frica. {W}e previously showed in vitro that {T}bg is able to excrete/secrete a large number of proteins, including {T}ranslationally {C}ontrolled {T}umor {P}rotein ({TCTP}). {M}oreover, the tctp gene was described previously to be expressed in {T}bg-infected flies. {A}side from its involvement in diverse cellular processes, we have investigated a possible alternative role within the interactions occurring between the trypanosome parasite, its tsetse fly vector, and the associated midgut bacteria. {I}n this context, the {T}bg tctp gene was synthesized and cloned into the baculovirus vector p{A}c{GHLT}-{A}, and the corresponding protein was produced using the baculovirus {S}podoptera frugicola (strain 9) / insect cell system. {T}he purified recombinant protein r{T}bg{TCTP} was incubated together with bacteria isolated from the gut of tsetse flies, and was shown to bind to 24 out of the 39 tested bacteria strains belonging to several genera. {F}urthermore, it was shown to affect the growth of the majority of these bacteria, especially when cultivated under microaerobiosis and anaerobiosis. {F}inally, we discuss the potential for {TC} {IP} to modulate the fly microbiome composition toward favoring trypanosome survival.}, keywords = {bacteria midgut flora ; {G}lossina ; {T}rypanosoma brucei gambiense ; {T}ranslationally {C}ontrolled {T}umor {P}rotein ({TCTP}) ; {AFRIQUE} {SUBSAHARIENNE}}, booktitle = {}, journal = {{G}ut {M}icrobes}, volume = {8}, numero = {5}, pages = {413--427}, ISSN = {1949-0976}, year = {2017}, DOI = {10.1080/19490976.2017.1331833}, URL = {https://www.documentation.ird.fr/hor/fdi:010071510}, }
