@article{fdi:010071347, title = {{T}emporal and spatial expression of genes involved in {DNA} methylation during reproductive development of sexual and apomictic {E}ragrostis curvula}, author = {{S}elva, {J}. {P}. and {S}iena, {L}. and {R}odrigo, {J}. {M}. and {G}arbus, {I}. and {Z}appacosta, {D}. and {R}omero, {J}. {R}. and {O}rtiz, {J}. {P}. {A}. and {P}essino, {S}. {C}. and {L}eblanc, {O}livier and {E}chenique, {V}.}, editor = {}, language = {{ENG}}, abstract = {{R}ecent reports in model plant species have highlighted a role for {DNA} methylation pathways in the regulation of the somatic-to-reproductive transition in the ovule, suggesting that apomixis (asexual reproduction through seeds) likely relies on {R}d{DM} downregulation. {O}ur aim was therefore to explore this hypothesis by characterizing genes involved in {DNA} methylation in the apomictic grass {E}ragrostis curvula. {W}e explored floral transcriptomes to identify homologs of three candidate genes, for which mutations in {A}rabidopsis and maize mimic apomixis ({A}t{AGO}9/{Z}m{AGO}104, {A}t{CMT}3/{Z}m{DMT}102/{Z}m{DMT}105, and {A}t{DDM}1/{Z}m{CHR}106), and compared both their spatial and temporal expression patterns during reproduction in sexual and apomictic genotypes. {Q}uantitative expression analyses revealed contrasting expression patterns for the three genes in apomictic vs sexual plants. {I}n situ hybridization corroborated these results for two candidates, {E}c{AGO}104 and {E}c{DMT}102, and revealed an unexpected ectopic pattern for the {AGO} gene during germ line differentiation in apomicts. {A}lthough our data partially support previous results obtained in sexual plant models, they suggest that rather than an {R}d{DM} breakdown in the ovule, altered localization of {A}t{AGO}9/{Z}m{AGO}104 expression is required for achieving diplospory in {E}. curvula. {T}he differences in the {R}d{DM} machinery acquired during plant evolution might have promoted the emergence of the numerous apomictic paths observed in plants.}, keywords = {}, booktitle = {}, journal = {{S}cientific {R}eports - {N}ature}, volume = {7}, numero = {}, pages = {art. 15092 [11 p.]}, ISSN = {2045-2322}, year = {2017}, DOI = {10.1038/s41598-017-14898-5}, URL = {https://www.documentation.ird.fr/hor/fdi:010071347}, }
