@article{fdi:010069396, title = {{M}olecular survey of {D}irofilaria immitis and {D}irofilaria repens by new real-time {T}aq{M}an ({R}) {PCR} assay in dogs and mosquitoes ({D}iptera : {C}ulicidae) in {C}orsica ({F}rance)}, author = {{T}ahir, {D}. and {B}ittar, {F}. and {B}arre-{C}ardi, {H}. and {S}ow, {D}. and {D}ahmani, {M}. and {M}ediannikov, {O}leg and {R}aoult, {D}. and {D}avoust, {B}. and {P}arola, {P}.}, editor = {}, language = {{ENG}}, abstract = {{D}irofilaria immitis and {D}. repens are filarioid nematodes of animals and humans, transmitted by the bite of infected mosquitoes. {D}omestic and wild canids are a major natural host and reservoir for these parasites. {I}n this study, we designed a duplex real-time {PCR} protocol targeting the mitochondrial cytochrome c oxidase subunit {I} ({COI}) gene, detecting both {D}. immitis and {D}. repens using two primer pairs and two {D}irofilaria-specific hydrolysable probes. {T}he sensitivity and specificity of the primers and probes were tested in both experimental and naturally infected samples. {T}he detection limits of this assay were evaluated using plasmid {DNA} from {D}. immitis and {D}. repens. {N}o cross-reaction was observed when testing this system against {DNA} from other filarial nematodes. {T}he detection limit of the real-time {PCR} system was one copy per reaction mixture containing 5 {I} of template {DNA}. {F}ield application of the new duplex real-time assay was conducted in {C}orsica. {T}he prevalence rate of {D}. immitis was 21.3% (20/94) in dogs. {I}n a locality where most dogs with {D}irofilaria spp. infection were found, {D}. immitis and {D}. repens were detected in 5% (20/389) and 1.5% (6/389) of the {A}edes albopictus population, respectively. {T}hese results suggest that this sensitive assay is a powerful tool for monitoring dirofilariosis in endemic or high risk areas.}, keywords = {{FRANCE} ; {CORSE}}, booktitle = {}, journal = {{V}eterinary {P}arasitology}, volume = {235}, numero = {}, pages = {1--7}, ISSN = {0304-4017}, year = {2017}, DOI = {10.1016/j.vetpar.2017.01.002}, URL = {https://www.documentation.ird.fr/hor/fdi:010069396}, }
