@article{fdi:010069288, title = {{U}se of eschar swabbing for the molecular diagnosis and genotyping of {O}rientia tsutsugamushi causing scrub typhus in {Q}uang {N}am province, {V}ietnam}, author = {{V}iet, {N}. {L}. and {L}aroche, {M}. and {P}ham, {H}. {L}. {T}. and {V}iet, {N}. {L}. and {M}ediannikov, {O}leg and {R}aoult, {D}. and {P}arola, {P}.}, editor = {}, language = {{ENG}}, abstract = {{B}ackground {S}crub typhus is a rickettsiosis which is caused by {O}rientia tsutsugamushi and occurs throughout the {A}sia-{P}acific region. {M}olecular diagnosis of rickettsioses using eschar swabs has recently emerged, and may be very useful for the diagnosis of these diseases in tropical settings. {M}ethodology/{P}rincipal findings {Q}uantitative polymerase chain reaction (q{PCR}) was used to detect {O}. tsutsugamushi {DNA} in whole blood and eschar swab specimens of 67 patients who were clinically suspected of scrub typhus in {Q}uang {N}am province, {V}ietnam. {A}mong the 20 patients for whom both eschar and whole blood were obtained, 17 (85%) of the eschar specimens and 5 (25%) of the whole blood specimens tested positive for {O}. tsutsugamushi. {G}enetic analysis of the 56-k{D}a {TSA} gene sequences demonstrated that the 14 sequences obtained in this study, including 12 eschar swabs and 2 whole blood specimens, were related to 4 groups: {K}arp, {K}awasaki, {G}illiam ({JG}-v and {TG}-v) and {TA}716. {T}he majority (9/14; 64.4%) of contemporary {O}. tsutsugamushi genotypes in {Q}uang {N}am province were related to the {K}arp group. {C}onclusions {T}hese results suggest that polyclonal antigen pools used for serological testing in the future should contain at least {K}arp, {K}awasaki, {G}illiam and {TA}716 antigens for {V}ietnamese patients, as well as patients who have traveled to {V}ietnam. q{PCR} after eschar swabbing should be considered for molecular diagnosis of scrub typhus in endemic patients as well as in travelers, since it is easy to perform and appears very useful for the rapid detection of {O}rientia tsutsugamushi in the early phase of infection.}, keywords = {{VIET} {NAM}}, booktitle = {}, journal = {{P}los {N}eglected {T}ropical {D}iseases}, volume = {11}, numero = {3}, pages = {e0005397 [16 p.]}, ISSN = {1935-2735}, year = {2017}, DOI = {10.1371/journal.pntd.0005397}, URL = {https://www.documentation.ird.fr/hor/fdi:010069288}, }
